The aim of this project was to select highly sensitive and specific aptamer capable of detecting Fusarium graminearum (Fg) and F. pseudograminearum (Fp) species. The aptamer-based testing allows early detection of fungal infection in the field. Fungi are known to change their activity and may often undergo genetic changes. The classic "gene for gene" theory explains the relationship between host plant cultivars and fungal pathogenic races.

A database consisting of 23 tripeptides was used to study the quantitative relationships between electric surface potential descriptors and antioxidant activity QESARs. The important structural descriptors SaaNH_acnt, SsOH_acnt, SaaN, SaaN_acnt, SsssCH, SaaaC, SsNH3p, SdO, SdO_acnt were selected for constructing the linear models QESARs with genetic algorithm. The best 4-variable linear model QESARlinear including the structural descriptors SaaN, SdO, SdO_acnt and SsOH_acnt was constructed. The quality QESARlinear was exhibited in statistical values R2 fitness of 97.

Breast cancer (BC), the most common type of cancer among women worldwide, is a polygenetic disease which is caused by the interaction of several genes. Understanding the genetic factors for early diagnosis of BC is crucial to ensure the survival of BC patients. MicroRNA 27a (miR-27a), an oncogenic miRNA, has been predicted to target on the tumor suppressor ZBTB10 that can regulate many processes of cell.

P-glycoprotein (Pgp), a membrane pump often responsible for the multidrug resistance of cancer cells, undergoes conformational changes in the presence of substrates/modulators, or upon ATP depletion, reflected by its enhanced reactivity with the UIC2 monoclonal antibody. When the UIC2-shift was elicited by certain modulators (e.g. cyclosporin A or vinblastine, but not with verapamil or Tween 80), the subsequent binding of other monoclonal anti-Pgp Ig sharing epitopes with UIC2 (e.g. MM12.10) was abolished [Nagy, H., Goda, K., Arceci, R., Cianfriglia, M., ´ Mechetner, E. & Szabo Jr, G.

A cytosolic thyroid-hormone-binding protein (xCTBP), predominantly responsible for the major binding activity of T3 in the cytosol of Xenopus liver, has been shown to be identical to aldehyde dehydrogenase class 1 (ALDH1) [Yamauchi, K., Nakajima, J., Hayashi, H., Horiuchi, R. & Tata, J.R. (1999) J. Biol. Chem. 274, 8460–8469]. Within this paper we surveyed which signaling, and other, compounds affect the thyroid hormone binding activity and aldehyde dehydrogenase activity of recombinant Xenopus ALDH1 (xCTBP/xALDH1) while examining the relationship between these two activities....

The ethanol utilization pathway inAspergillus nidulansis a model system, which has been thoroughly elucidated at the biochemical, genetic and molecular levels. Three main ele-ments are involved: (a) high level expression of the positively autoregulated activator AlcR; (b) the strong promoters of the structural genes for alcohol dehydrogenase (alcA)and aldehyde dehydrogenase (aldA); and (c) powerful activa-tion of AlcRby the physiological inducer, acetaldehyde, produced from growth substrates such as ethanol and L-threonine. ...

The RNA recognition motif (RRM), also known as RNA-binding domain (RBD) or ribonucleoprotein domain (RNP) is one of the most abundant protein domains in eukaryotes. Based on the comparison of more than 40 structures including 15 complexes (RRM–RNA or RRM–protein), we reviewed the structure–function relationships of this domain. We identified and classified the different structural elements of the RRM that are import-ant for binding a multitude of RNA sequences and proteins.

NAD + -dependent (R)-2-hydroxyglutarate dehydrogenase (HGDH) cata-lyses the reduction of 2-oxoglutarate to (R)-2-hydroxyglutarate and belongs to the D-2-hydroxyacid NAD + -dependent dehydrogenase (D-2-hydroxyacid dehydrogenase) protein family. Its crystal structure was determined by phase combination to 1.98 A˚ resolution. Structure–function relationships obtained by the comparison of HGDH with other members of theD-2-hydroxyacid dehydrogenase family give a chemically satisfying view of the substrate stereoselectivity and catalytic requirements for the hydride trans-fer reaction.

The efficient engineering of enzymes with novel activities remains an ongoing challenge. Towards this end, genetic selection techniques provide a method for finding rare solutions to catalytic problems that requires only a limited foreknowledge of structure–function relationships. We have used genetic selections to extensively probe the structure and mechanism of chorismate mutases. The insights gained from these investigations will aid future enzyme design efforts.

Mitochondrial fatty acid oxidation deficiencies are due to genetic defects in enzymes of fatty acidb-oxidation and transport proteins.Genetic defects have been identified in most of the genes where nearly all types of sequence vari-ations (mutation types) have beenassociatedwithdisease. In this paper, we will discuss the effects of the various types of sequence variations encountered and review current know-ledge regarding the genotype–phenotype relationship, espe-cially in patients with acyl-CoA dehydrogenase deficiencies where sufficient material exists for a meaningful discussion....

Huntington’s disease (HD) is a progressive neurodegenerative disorder characterized by multifarious dysfunctional alterations including mitochon-drial impairment. In the present study, the formation of inclusions caused by the mutation of huntingtin protein and its relationship with changes in energy metabolism and with pathological alterations were investigated both in transgenic and 3-nitropropionic acid-treated mouse models for HD.
