Free amino acids are found in spermatozoa and seminal plasma. Their qualitative and quantitative presence have been affected by many factors. Under in vitro storage conditions, their effects on spermatozoa in animal species and also human have been concerned over the last decades. In this study, the effects of tryptophan, phenylalanine and cysteine on DNA integrity, lipid peroxidation, viability and motility parameters in spermatozoa of the common carp (Cyprinuscarpio) were assessed during short-term storage.

The effects of UV-B light force, exposure time and incubation period on producing caffeic acid derivatives and growth parameters in Echinacea purpurea callus and suspension culture were assessed. UV-B led to an increment of all growth parameters and antioxidant activity in callus and cell suspension and caffeic acid derivatives in cell suspension by increasing incubation period.

The influence of different parameters on IAA production of these strains was evaluated by the “one factor at a time” method and the obtained results showed that the best conditions for IAA production of DTAN1-M5 were a medium with the following compositions: 1 % peptone, 0.5 % sucrose, 0.6 % yeast extract, 0.5 % NaCl, pH 7.1, supplemented with 0.2 mg/mL L-tryptophan, which was incubated in a shaker at 150 rpm and 30 oC for 84 hours. The maximum amount of IAA was 84.13 µg/mL.

Cellulolytic bacteria were previously isolated from the termite guts and efforts were taken to optimize parameters for cellulase production. Activities of diversity of enzymes which include, CMCase, avicelase and filterpaperase (FPase) were studied from crude cellulase extracts. CMCase activity was found optimum at different temperatures (35°C, 40°C and 45°C) at pH 7 and after 48 hrs of incubation time.

Amylase producing thermophilic bacterial strain was isolated from Jeori hot water spring, Rampur Bushahr of Shimla district of Himachal Pradesh, India and identified as Bacillus thuringiensis J2 using 16S rRNA gene sequencing and deposited in NCBI gene bank vide accession number |KY990713|. Medium components and process parameters were optimized with respect to substrate concentration, carbon sources, nitrogen sources, incubation period, pH and temperature for an enhanced amylase production.

Mycelial growth and exopolysaccharide production of Fomitopsis feei was studied in common production medium by changing different physiological parameters such as carbon, nitrogen sources, inoculum size, incubation time, pH, temperature, agitation. Glucose, ammonium sulphate, 6mm of inoculum, 8th and 14th days of incubation, pH 3, 30°C temperature and 150 rpm agitation were the best for the production of mycelium.

This study was aimed to investigate the potential of single culture of fungi Aspergillus niger, to decolorize reactive dyes from synthetic solution. Different parameters such as pH, time, temperature, agitation rate and various carbon, nitrogen and inorganic salts source, were optimized for decolorization of reactive blue 19 and reactive black 5 dyes. Aspergillus niger showed maximum dye decolorization under optimum condition and found to be more efficient when added in the dye solution of pH 8 and 10 with agitation at 130 rpm and incubation time for 7 days with 25°C.

Deproteinated cheese whey was used for biomass production of probiotic strain Lactobacillus helveticus MTCC 5463 in a biofermenter. Optimization of growth parameters such as temperature, pH and time of incubation as well as nutrient supplementation of cheese whey was carried out using response surface methodology (RSM). The chickpea genotype ICCV 5530, which is moderately resistant and ICCV 4991, susceptible to Ascochyta rabiei were selected to identify the changes in biochemical activity in response to infection by the fungus Ascochyta rabiei. Twelve day old seedlings were inoculated with spore suspension of the pathogen at 5 × 104 spore/ml concentration using a Haemocytometer and incubated under Controlled Environmental Facility (CEF). Sampling was done at 2, 4 and 8 days after inoculation.

Incubation period as a measure of infection efficiency for isolates of leaf blast pathogen in pearl millet and rice is an important parameter to testify host specificity as well as resistance against the pathogen. Incubation period (IP50) for leaf blast symptom development on pearl millet and rice using respective isolates (pearl millet isolate on pearl millet and rice isolate on rice) were estimated to be 4.98 and 6.23 days respectively at 26±1°C.

Bacillus subtilis has been widely used to produce hydrolytic enzymes for various industrial applications. In this study, solid state fermentation was carried out using various agro- industrial wastes for the production of alpha amylase using B. subtilis. Among the tested agro-residues, wheat bran supported more enzyme production. The process parameters elucidated were type of substrate, incubation time, pH, moisture content of the medium, carbon source and nitrogen source.
