This dissertation and project analyse the ongoing crisis in news journalism and evidencebased inquiries. Through practice-led research, the completed prototype Labyrinths & Leaks examines methods for overlapping evidence through the interactive documentary (i-doc) form. This research examines what happens when we break apart narrative sequencing. Rather than a gradual narrowing of the linear storyline, there is a network of openings and opportunities afforded through the i-doc form.

Correct application of the classical factorial F-test depends on normality and homogeneity of variance assumptions. If these assumptions are violated the type I error rate will be inflated and power of the test will be decreased. Therefore nonparametric statistical tests have been proposed to analyze the interaction effects in factorial designs.

The design of b-glycosidases with planed substrate specificity for biotechnological application has received little attention. This is mostly a consequence of the lack of data on the molecular basis of the b-glycosidase specificity, namely data on the energy of the noncovalent interactions in the enzymetransition state complex. In an attempt to fill this gap, sitedirected mutagenesis and enzyme steady-state kinetic experiments with different substrates were conducted, using as model a digestive b-glycosidase (glycoside hydrolase family 1) from Spodoptera frugiperda (Lepidoptera) (Sfbgly50).

We report here the first crystal structure of a stable isosteric analogue of 1,3-bisphospho-D-glyceric acid (1,3-BPGA) bound to the catalytic domain of Trypanosoma cruzi glycosomal glyceraldehyde-3-phosphate dehydrogenase (gGAPDH)in which the two phosphoryl moieties interact with Arg249. This complex possibly illustrates a step of the catalytic process by which Arg249 may induce compression of the product formed, allowing its expulsion fromthe active site.

ANKHD1 proteins are known to possess multiple ankyrin repeat domains and a single KH domain with no known function. Using yeast two-hybrid system analysis, we identified a novel splice variant of ANKHD1. The interaction between cell-adhesion molecules CD2 and CD58 is critical for an immune response. Modulation or inhibition of these interactions has been shown to be thera-peuticallyuseful. Synthetic 12-mer linear and cyclic peptides, and cyclic hexapeptides based on rat CD2 protein, were designed tomodulateCD2–CD58 interaction.
