The aim of this work was to screen and validate biomarkers of ovarian cancer-initiating cells to detect the mechanisms of recurrence of epithelial ovarian cancer (EOC). Methods: Stably labelled the amino acid in side population (SP) cells of epithelial ovarian cancer which were rich in cancer-initiating cells and non-SP cells with isotope in culture and differentially expressed cellular membrane proteins in SP cells were identified through proteomics technology.

We evaluate here different analytical strategies for the chromatographic separation and determination of N-acetyl-5-methoxytryptamine (MEL) and its oxidative metabolites N1-acetyl-N2-formyl-5- methoxykynuramine (AFMK), N1-acetyl-5-methoxykynuramine (AMK) and cyclic 3-hydroxymelatonin (c3OHM) in cell culture samples.

Cancerous cells can recycle metabolic ammonium for their growth. As this ammonium has a low nitrogen isotope ratio (15N/14N), its recycling may cause cancer tissue to have lower 15N/14N than surrounding healthy tissue. We investigated whether, within a given tissue type in individual mice, tumoral and healthy tissues could be distinguished based on their 15N/14N.

Tracing stable isotopes, such as 13C using various mass spectrometry (MS) methods provides a valuable information necessary for the study of biochemical processes in cells. However, extracting such information requires special care, such as a correction for naturally occurring isotopes, or overlapping mass spectra of various components of the cell culture medium.

Modelling of the fluxes in central metabolism can be performed by combining labelling experiments with metabolite balancing. Using this approach, multiple samples from a cultivation of Saccharomyces cerevisiae in metabolic and isotopic steady state were analysed, and the metabolic fluxes in central metabolism were estimated. In the various samples, the estimates of the central metabolic pathways, the tricarboxylic acid cycle, the oxidative pentose phosphate pathway and the anaplerotic pathway, showed an unprecedented reproducibility....

To shed light on the mechanism of isotopic exchange of a-protons in amino acids catalyzed by pyridoxal phosphate (PLP)-dependent enzymes, we studied the kinetics of quinonoid intermediate formation for the reactions of tyrosine phenol-lyase with L-phenylalanine, L-methionine, and theira-deuterated analogues inD2O, andwe compared the results with the rates of the isotopic exchange under the same conditions.Wehave found that, in theL-phenylalanine reaction, the internal returnof thea-proton is operative, and allowing for its effect, the exchange rate is accounted for satisfactorily....

The introduction of noncanonical amino acids and bio-physical probes into peptides and proteins, and total or segmental isotopic labelling has the potential to greatly aid thedeterminationofproteinstructure, functionandprotein– protein interactions. To obtain a peptide as large as possible by solid-phase peptide synthesis, native chemical ligation was introduced to enable synthesis of proteins of up to 120 amino acids in length.

7-Ethoxy (OEt) coumarin has been used as a model substrate in many cyto-chrome P450 (P450) studies, including the use of kinetic isotope effects to probe facets of P450 kinetics. P450s 1A2 and 2E1 are known to be the major catalysts of 7-OEt coumarinO-deethylation in human liver micro-somes.

A platform based on hydrophilic interaction chromatography in combina-tion with Fourier transform mass spectrometry was developed in order to carry out metabonomics of Drosophila melanogasterstrains. The method was able to detect230 metabolites, mainly in the positive ion mode, after checking to eliminate false positives caused by isotope peaks, adducts and fragment ions.

This book explores the diversity of topics related to radiometric dating, with particular emphasis on the method of radiocarbon dating and a cross-check of its results with luminescence measurements. Chemistry and the global carbon cycle is an essential element of life on Earth, because carbon is a component of every living cell, plants and animals. The radioactive isotope 14C was found useful for dating, for which in 1960 Willard Frank Libby received the Nobel Prize in Chemistry.

Approach to the Patient: Polycythemia As shown in Fig. 58-18, the first step is to document the presence of an increased red cell mass using the principle of isotope dilution by administering 51 Cr-labeled autologous red blood cells to the patient and sampling blood radioactivity over a 2-h period. If the red cell mass is normal (36 mL/kg in men, 32 mL/kg in women), serum EPO levels should be measured. If EPO levels are low or unmeasurable, the patient most likely has polycythemia vera.
