This study aimed to establish culture medium changing system to improve the efficiency of bovine somatic cell nuclear transfer protocol. Our results showed that using the culture medium of Takahashi and First 1992 produced the cloned bovine blastocyst at the highest rate (9.4%, P < 0.05) among different modified synthetic oviduct fluid (mSOF) media.

This polymeric ligand was reacted with PdCl 2 to obtain a polymeric Pd(0) complex. The TEM image of the Pd catalyst showed good dispersion of catalytic sites. The Pd catalyst exhibits excellent activity and stability in copper-free Sonogashira–Hagihara cross-coupling reactions under aerobic conditions. This protocol can be applied efficiently to the coupling reactions of chloro- as well as iodo- and bromo-arenes. The catalyst can be reused several times without any considerable decrease in its activity.

An efficient and facile method has been developed for the synthesis of alkyl bromides from various alcohols under mild conditions using a triphenylphosphine (PPh3)/1,2-dibromotetrachloroethane (DBTCE) complex in excellent yields and very short time (5 min). This method can also be applied for the transformation of chiral alcohols to their corresponding bromides in very high enantiomeric excess. The PPh3 /DBTCE complex is also successfully applied to ring-opening reactions of cyclic ethers in mild conditions.

This inhibits 3′ adapter ligation and makes library preparation particularly challenging. To reduce bias, the NEBNext kit (New England Biolabs) uses polyethylene glycol (PEG), the NEXTflex V2 kit (BIOO Scientific) uses both randomised adapters and PEG, and the novel SMARTer (Clontech) and CATS (Diagenode) kits avoid ligation altogether. Here we compared these methods with Illumina’s classical TruSeq protocol regarding the detection of normal and 2’ OMe RNAs. In addition, we modified the TruSeq and NEXTflex protocols to identify conditions that improve performance.
