MFG-E8 (milk fat globule-EGF factor 8) is a peripheral membrane glycoprotein, which is expressed abundantly in lactating mammary glands and is secreted in association with fat globules. This protein consists of two-repeated EGF-like domains, a mucin-like domain and two-repeated discoidinlike domains (C-domains), and contains an integrin-binding motif (RGD sequence) in the EGF-like domain. The intrachloroplastic localization of post-transcriptional gene expression steps represents one key determinant for the regulation of chloroplast development. We have character-ized an RNA binding protein of 63kDa (RBP63) from Chlamydomonas reinhardtiichloroplasts, which cofraction-ates with stromal thylakoid membranes. Solubility proper-ties suggest that RBP63is a peripheral membrane protein.

Peripheral nerve injury is normally followed by a robust regenerative response. Here we describe the early changes associated with injury from the initial rise in intracellular calcium and the subsequent activation of transcription factors and cytokines leading to an inflammatory reaction, and the expression of growth factors, cytokines, neuropeptides, and other secreted molecules involved in cell-to-cell communication promoting regen-eration and neurite outgrowth.

We report here thatEscherichia coli pyrophosphatase aggregates in the presence of millimolar Cd 2+ . This highly cooperative process was specific to both the metal ion and the protein and could be reversed fully by decreasing the Cd 2+ concentration. Aggregation was enhanced by Mg 2+ , the natural cofactor of pyrophosphatase, and Mn 2+ . Mutations at the intersubunit metal-binding site had no effect, whereas mutation at Glu139, which is part of the peripheral metal-binding site found in pyrophosphatase crystals near the contact region between two enzyme mole-cules, suppressed aggregation.

The divalent metal transporter (DMT1) is a 12-transmem-brane domain protein responsible for dietary iron uptake in the duodenum and iron acquisition from transferrin in peripheral tissues. The transmembrane domain 4 (TM4) of DMT1 has been shown to be crucial for its biological function. Here we report the 3D structure and topology of the DMT1-TM4 peptide by NMR spectroscopy with simulated annealing calculations in membrane-mimetic environments, e.g. 2,2,2-trifluoroethanol and SDS micelles.
