Two ORFs, cphA and cphB, encoding proteins CphA and CphB with strong similarities to plant phytochromes and to the cyanobacterial phytochrome Cph1 of Synechocystis sp. PCC 6803 have been identified in the filamentous cyanobacterium Calothrix sp. PCC7601. While CphA carries a cysteine within a highly conserved amino-acid sequence motif, to which the chromophore phytochromobilin is covalently bound in plant phytochromes, in CphB this position is changed into a leucine.

Subunit G of photosystem I is a nuclear-encoded protein, predicted to form two transmembrane a-helices separated by a loop region. We use in vitro import assays to show that the positively charged loop domain faces the stroma, whilst the N- and C-termini most likely face the lumen. PSI-G constructs in which a His- or Strep-tag is placed at the C-terminus or in the loop region insert with the same topology as wild-type photosys-tem I subunit G (PSI-G).

Replication protein A (RPA), a heterotrimeric protein composed of 70, 32 and 14-kDa subunits, has been shown to be essential for DNA replication, repair, recombination, and transcription. Previously, we found that, in two seed plants, rice and Arabidopsis, there are two different types of RPA70-kDa subunit. Substantial biochemical and genetic characterization of these two subunits, termed OsRPA70aand OsRPA70bor AtRPA70a and AtRPA70b, respectively, is described in this report.

The conduction of protons through human Nox2 has pre-viously been shown to be dependent upon His115. Align-ment of sequences for both animal and plant Nox proteins indicated that histidines 115 and 119 are both highly con-served, while His111 was conserved among animal homo-logues of Nox1–4. To investigate the possible role that these histidine residues might play in the conduction of protons through Nox2, we have introduced both paired and single mutations into these histidine residues.

During a search for cDNAs encoding plant sterol acyl-transferases, we isolated four full-length cDNAs fromAra-bidopsis thaliana that encode proteins with substantial identity with animal lecithin : cholesterol acyltransferases (LCATs). Volkensin, a type 2 ribosome-inactivating protein from the roots of Adenia volkensiiHarms (kilyambiti plant) was characterized both at the protein and nucleotide level by direct amino acid sequencing and cloning of the gene enco-ding the protein. A novel, type 1 ribosome-inactivating protein designated charybdin was isolated from bulbs of Charybdis maritimaagg. The protein, consisting of a single polypeptide chain with a molecular mass of 29 kDa, inhibited trans-lation in rabbit reticulocytes with an IC50of 27.2 nm.
