A large number of studies have demonstrated the wound-healing effects of LLLT in vitro, in animal models, and in clinical practice. However, there are also differences in the study results, which are dose and wavelength dependent of LLLT. Objective: Evaluation of the wound healing process in experimental animals treated with low- level laser therapy in clinical and histopathology.

Connective tissue growth factor/hypertrophic chondrocyte specific gene product 24 (CTGF/Hcs24) promotes proliferation and differentiation of chondrocytes in culture. We investigated the roles of two major types of mitogen activated protein kinase (MAPK) in the promotion of proliferation and differentiation by CTGF/Hcs24. Here we report the effects of the MAPKK/MEK 1/2 inhibitor, PD098059, and p38 MAPK inhibitor, SB203580, in a human chondrosarcoma-derived chondrocytic cell line (HCS-2/8) and rabbit growth cartilage (RGC) cells treated with CTGF/ Hcs24....

Conventional and saturation transfer electron paramagnetic resonance spectroscopy (EPR and ST EPR) was used to study the orientation of probe molecules in muscle fibers in different intermediate states of the ATP hydrolysis cycle. A separate procedure was used to obtain ST EPR spectra with precise phase settings even in the case of samples with low spectral intensity. Fibers prepared from rabbit psoas muscle were labeled with isothiocyanate spin labels at the reactive thiol sites of the catalytic domain of myosin....

Lamjapin,anoveltypeIribosome-inactivating protein, has been isolated from kelp (Laminaria japonicaA), a marine alga. This protein has been extensively purified through multiple chromatography columns. With a molecular mass of36 kDa, lamjapin is slightly larger than the other known single-chain ribosome-inactivating proteins from the higher plants. Lamjapin can inhibit protein synthesis in rabbit reticulocyte lysate with an IC50 of 0.69 nM.

Mammalian tubulins and actins attain their native con-formation following interactions with CCT (the cytosolic chaperonin containing t-complex polypeptide 1). To study the b-tubulin folding in lower eukaryotes, an isotype of b-tubulin (b-T1) from the Antarctic ciliate Euplotes focardii, was expressed in Escherichia coli. Folding analysis was performed by incubation of the 35 S-labeled, denatured b-T1 in the presence, or absence, of purified rabbit CCT and cofactor A, a polypeptide that stabilizes folded monomericb-tubulin. ...

A naturally occurring hemagglutinin was detected in the serum of the freshwater crab, Paratelphusa jacquemontii (Rathbun). Hemagglutination activity with different mam-malian erythrocytes suggested a strong affinity of the serum agglutinin for horse and rabbit erythrocytes. The most potent inhibitor of hemagglutination proved to be bovine submaxillary mucin. The lectin was purified by affinity chromatography using bovine submaxillary mucin-coupled agarose. The molecular mass of the purified lectin was 34 kDa as determined by SDS/PAGE....

Anandamide is an endogenous ligand for cannabinoid receptor and its protein-mediated transport across cellular membranes has been demonstrated in cells derived from brain as well as in cells of the immune system. This lipid is inactivated via intracellular degradation by a fatty acid amidohydrolase (FAAH). In the present study, we report that rabbit platelets, in contrast to human platelets, do not possess a carrier-mediated mechanism for the transport of [ 3 H]anandamide into the cell, i.e. cellular uptake was not temperature dependent and its accumulation was not satu-rable. ...

Atomic models of the myosin motor domain with different bound nucleotides have revealed the open and closed con-formationsof the switch2 element [Geeves,M.A.&Holmes, K.C. (1999)Annu. Rev. Biochem.68, 687–728]. The two conformations are in dynamic equilibrium, which is con-trolled by the bound nucleotide. In the present work we attempted to characterize the flexibilityof themotor domain in the open and closed conformations in rabbit skeletal myosin subfragment 1.

Vertebrate troponin regulates muscle contraction through alternative bind-ing of the C-terminal region of the inhibitory subunit, troponin-I (TnI), to actin or troponin-C (TnC) in a Ca 2+ -dependent manner. To elucidate the molecular mechanisms of this regulation by molluskan troponin, we com-pared the functional properties of the recombinant fragments of Akazara scallop TnI and rabbit fast skeletal TnI.

Studying the oxygenation kinetics of (19R⁄S,5Z,8Z,11Z,14Z)-19-hydroxy-eicosa-5,8,11,14-tetraenoic acid (19-OH-AA) by rabbit 15-lipoxygenase-1 we observed a pronounced oxygen dependence of the reaction rate, which was not apparent with arachidonic acid as substrate. Moreover, we found that peroxide-dependent activation of the lipoxygenase depended strongly on the oxygen concentration.

Comparison of the crystal structures of threeMicropechis ikahekaphos-pholipase A2 isoenzymes (MiPLA2, MiPLA3 and MiPLA4, which exhibit different levels of pharmacological effects) shows that their C-terminus (residues 110–124) is the most variable. M-Type receptor binding affinity of the isoenzymes has also been investigated and MiPLA4 binds to the rabbit M-type receptor with high affinity. Examination of surface charges of the isoenzymes reveals a trend of increase in positive charges with potency....

The effect of gadolinium ions on the sarcoplasmic reticulum (SR) calcium release channel⁄ryanodine receptor (RyR1) was studied using heavy SR (HSR) vesicles and RyR1 isolated from rabbit fast twitch muscle. In the [ 3 H]ryanodine binding assay, 5lmGd 3+ increased the Kd of the [ 3 H]ry-anodine binding of the vesicles from 33.8 nmto 45.6 nmwhileBmax, refer-ring to the binding capacity, was not affected significantly. In the presence of 18 nm[ 3 H]ryanodine and 100lmfree Ca 2+ ,Gd 3+ inhibited the binding of the radiolabeled ryanodine with an apparentKd value of 14.

A protocol to follow the processing of angiotensin I into angiotensin II by rabbit angiotensin-converting enzyme (ACE) and its inhibition by a novel natural antagonist, the leech osmoregulator factor (LORF) using capillary zonal electrophoresis is described. The experiment was carried out using the Beckman PACE system and steps were taken to determine (a) the migration profiles of angiotensin and its yielded peptides, (b) the minimal amount of angiotensin II detected, (c) the use of different electrolytes and (d) the concentration of inhibitor. ...

In order to determine the amino-terminal sequence requirements for protein N-myristoylation, site-directed mutagenesis of the N-terminal region was performed using tumor necrosis factor (TNF) mutants as model substrate proteins. Subsequently, the susceptibility of thesemutants to protein N-myristoylation was evaluated by metabolic labe-ling in anin vitrotranslation systemusing rabbit reticulocyte lysate. A TNF mutant having the sequence MGAAAAA AAA at its N-terminus was used as the starting sequence to identify elements critical for protein N-myristoylation....
