To learn about the correlation between allostery and ligand binding of the Tet repressor (TetR) we analyzed the effect of mutations in the DNA read-ing head–core interface on the effector specific TetR i2 variant. The same mutations in these subdomains can lead to completely different activities, e.g. the V99G exchange in the wild-type leads to corepression by 4-ddma-atc without altering DNA binding. However, in TetR i2 it leads to 4-ddma-atc dependent repression in combination with reduced DNA binding in the absence of effector. ...

The gene encoding an integrase of Mason–Pfizer monkey virus (M-PMV) is located at the 3¢-end of the pol open reading frame. The M-PMV integ-rase has not been previously isolated and characterized. We have now cloned, expressed, isolated, and characterized M-PMV integrase and com-pared its activities and primary structure with those of HIV-1 and other retroviral integrases. M-PMV integrase prefers untranslated 3¢-region-derived long-terminal repeat sequences in both the 3¢-processing and the strand transfer activity assays.

An open reading frame with fatty acid desaturase similarity was identified in the genome ofTrypanosoma brucei. The 1224 bp sequence specifies a proteinof 408 amino acids with 59% and 58% similarity toMortierella alpinaandArabid-opsis thalianaD12 desaturase, respectively, and 51% with A. thalianax3 desaturases. The histidine tracks that com-pose the iron-binding active centers of the enzyme were more similar to those of thex3 desaturases.

The genome of the mesophilic Gram-negative bacteriumBurkholderia thai-landensis contains an open reading frame (i.e. theBth_I1158gene) that has been annotated as a putative ribokinase and PFK-B family member. Nota-bly, although the deduced amino acid sequence of the gene showed only 29% similarity to the recently identified nucleoside kinase from hyperthermophilic archaeaMethanocaldococcus jannaschii, 15 of 17 residues reportedly involved in the catalytic activity of M.

NifS-like cysteine desulfurases are widespread enzymes involved in the mobi-lization of sulfur from cysteine. The genome of the filamentous diazotrophic cyanobacterium AnabaenaPCC 7120 contains four open reading frames potentially encoding NifS-like proteins. One of them, alr2505, belongs to the pkn22operon, which enablesAnabaenato cope with oxidative stress.
