Mitochondrial functional and structural impairment and generation of oxidative stress have been implicated in aging, various diseases and chemotherapies. This study analyzed azidothymidine (AZT)-caused failures in mitochondrial functions, in redox regulation and activation of the HIV-1 gene expression. We monitored intracellular concentrations of ATP and glutathione (GSH) as the indicators of energy production and redox conditions, respectively, during the time-course experiments with U937 and MOLT4 human lymphoid cells in the presence of AZT (0.05 mgÆmL)1) or H2O2 (0.01 mM) for 15–25 days.

Cloning and over-expression of human glucose 6-phosphate dehydrogenase (Glc6P dehydrogenase) has for the first time allowed a detailed kinetic study of a preparation that is genetically homogeneous and in which all the protein molecules are of identical age. The steady-state kinetics of the recombinant enzyme, studied by fluorimetric initial-rate measurements, gave converging linear Lineweaver–Burk plots as expected for a ternary-complex mechanism.

It is based on the assumption that cells developed optimal adaptation strategies to changing environmental conditions. In this study, we have characterized the influence of hypoxia on the expression of hydroxylases crucially involved in col-lagen fiber formation, such as prolyl-4-hydroxylases (Ph4) and procollagen lysyl-hydroxylases (PLOD). Using the rat vascular smooth muscle cell line A7r5, we found that an hypoxic atmosphere caused a characteristic time-dependent five- to 12-fold up-regulation of the mRNAs of the two P4h a-subunits [aI (P4ha1) andaII (P4ha2)] and of two lysyl-hydroxylases (PLOD1 and PLOD2).

For a long time, the haemerythrin family of proteins was considered to be restricted to only a few phyla of marine invertebrates. When analysing dif-ferential protein expression in the methane-oxidizing bacterium, Methylo-coccus capsulatus(Bath), grown at a high and low copper-to-biomass ratio, respectively, we identified a putative prokaryotic haemerythrin expressed in high-copper cultures.

Thebform of human cAMP-dependent protein kinase inhibitor (human PKIb), a novel heat-stable protein, was isolated with high yield using a bacterial expression system. Assays of PKI activity demonstrated that purified PKIb inhibits the catalytic subunit of cAMP-dependent protein kinase. FTIR, Raman spectroscopy and CD experiments implied that human PKIbcontained only small amounts of a-helix andb-structures, but large amounts of random coil and turn structures, which may explain its high thermosta-bility.

A generic sample normalization method applicable in relative comparison of mRNAs quantified with real-time polymerase chain reaction (PCR) is proposed. The method was applied in samples obtained from tomato seeds after osmopriming and aging treatments and from untreated seeds at early imbibition stage, when seeds had not completed germination.

Control analysis of the glycolytic flux was carried out in two fast-growth tumor cell types of human and rodent origin (HeLa and AS-30D, respect-ively). Determination of the maximal velocity (Vmax) of the 10 glycolytic enzymes from hexokinase to lactate dehydrogenase revealed that hexokin-ase (153–306 times) and phosphfructokinase-1 (PFK-1) (22–56 times) had higher over-expression in rat AS-30D hepatoma cells than in normal freshly isolated rat hepatocytes.

The mechanism behind the growth-promoting effect of insulin is a subject of debate. Employing RT4 bladder cancer cells, we examined the cross-talk between insulin and the epidermal growth factor system. We found that insulin induced a time- and dose-dependent (25–1000 nmolÆL )1 insulin) increase in mRNA expression of three ligands from the epidermal growth factor system.
