  • The thesis studied the development and implementation of the conceptual Continuous Improvement Teamworking (CIT) Model for strategic performance improvement in retail banking operations. The investigation using of a dominant qualitative to a Less-dominant quantitative approach as a mixed-method strategy revealed the importance of the CIT Model realised through the CTIO (Concern-Task-Interaction-Outcome) Problem-Solving Cycle. The CTIO Cycle is a variant form of the traditional Deming-Shewhart PDCA (Plan-Do-Check-Action) Cycle.

  • Biotherapeutics (BTs), one of the fastest growing classes of drug molecules, offer several advantages over the traditional small molecule pharmaceuticals because of their relatively high specificity, low off-target effects, and biocompatible metabolism, in addition to legal and logistic advantages. However, their clinical utility is limited, among other things, by their high immunogenic potential and/or variable therapeutic efficacy in different patient populations.

  • A Bayesian approach with frequentist validity has been developed to support inferences derived from a BLevel A^ in vivo-in vitro correlation (IVIVC). Irrespective of whether the in vivo data reflect in vivo dissolution or absorption, the IVIVC is typically assessed using a linear regression model. Confidence intervals are generally used to describe the uncertainty around the model. While the confidence intervals can describe population-level variability, it does not address the individual-level variability.

  • This review describes studies of particular enzymatically catalyzed reactions to investigate the possibility that catalysis is mediated by protein dynamics. That is, evolution has crafted the protein backbone of the enzyme to direct vibrations in such a fashion to speed reaction. The review presents the theoretical approach we have used to investigate this problem, but it is designed for the nonspecialist.

  • Modelling of the fluxes in central metabolism can be performed by combining labelling experiments with metabolite balancing. Using this approach, multiple samples from a cultivation of Saccharomyces cerevisiae in metabolic and isotopic steady state were analysed, and the metabolic fluxes in central metabolism were estimated. In the various samples, the estimates of the central metabolic pathways, the tricarboxylic acid cycle, the oxidative pentose phosphate pathway and the anaplerotic pathway, showed an unprecedented reproducibility....

  • The decoy approach against nuclear factor kB (NF-kB) is a useful tool to alter NF-kB dependent gene expression using synthetic oligonucleotides (ODNs) carrying NF-kB specific cis-elements. Unfortunately, ODNs are not stable and need to be extensively modified to be used in vivo or ex vivo. We have previously evaluated the possible use of peptide nucleic acids (PNAs) as decoy molecules. The backbone of PNAs is composed of N-(2-aminoethyl)glycine units, rendering these molecules resistant to both nucleases and proteases.

  • In seminiferous tubules, Sertoli cells provide structural and nutritional support for the developing germinal cells. Cellto-cell signaling and cell adhesion require proteoglycans expressed at the cell membrane. A preliminary biochemical and structural approach indicated that cell surface proteoglycans are mostly heparan sulfate proteoglycans (HSPG). Glypican-1, syndecans-1 and -4 were identified using a molecular approach.

  • Chlamydomonas reinhardtii, a unicellular green alga, contains a hydrogenase enzyme, which is induced by anaerobic adaptation of the cells. Using the suppression subtractive hybridization (SSH) approach, the differential expression of genes under anaerobiosis was analyzed. A PCR fragment with similarity to the genes of bacterial Fe-hydrogenases was isolated and used to screen an anaerobic cDNA expression library of C. reinhardtii.

  • For the biosynthesis of punicic acid (18:3 D9Z,11E,13Z )a (11,14)-linoleoyl desaturase activity has been proposed. To isolate this acyl-lipid-desaturase, PCR-based cloning was used. This approach resulted in the isolationof twocomplete cDNAs. The first isolated full-length cDNA harbors a sequence of 1350 bp encoding a protein of 395 amino acids. The second cDNA was 1415 bp long encoding a protein of 387 amino acids.

In this oxidase the electron donor, cytochromec, is covalently bound to subunit II of the cytochrome c oxidase. Oxidative electrochemical redox titrations in the visible spectral range yielded a midpoint potential of )0.01 ± 0.01 V (vs. Ag/AgCl/3MKCl, 0.218 V vs. SHE¢) for the hemec.

  • In this paper we address the problem of extracting features relevant for predicting protein±protein interaction sites from the three-dimensional structures of protein complexes. Our approach is based on information about evolutionary con-servation and surface disposition. We implement a neural network based system, which uses a cross validation proce-dure and allows the correct detection of 73% of the residues involved in protein interactions in a selected database comprising 226 heterodimers.

  • A new approach of predicting structural classes of protein domain sequences is presented in this paper. Besides the amino acid composition, the composition of several dipeptides, tripeptides, tetrapeptides, pentapeptides and hexapeptides are taken into account based on the step-wise discriminant analysis. The result of jackknife test shows that this new approach can lead to higher pre-dictive sensitivityand specificityfor reduced sequence similaritydatasets.
