To gain insight in the subcellular localization of tumor necrosis factor receptor-associated factor (TRAF4) we analyzed GFP chimeras of full-length TRAF4 and various deletionmutants derived thereof. While TRAF4–GFP (T4– GFP) was clearlylocalized in the cytoplasm, the N-terminal deletion mutant, T4(259–470), comprising the TRAF domain of the molecule, and a C-terminal deletion mutant consisting mainlyof the RING and zinc finger domains of TRAF4 were both localized predominantlyto the nucleus....

Our laboratory showed that bikunin, aKunitz-type protease inhibitor, suppresses 4b-phorbol 12-myristate 13-acetate (PMA)- or tumor necrosis factor-alpha (TNFa)-induced urokinase-type plasminogen activator (uPA) expression in different cell types. In addition to its effects on protease inhibition, bikunincouldbemodulatingother cellular events associated with the metastatic cascade.

Double-stranded RNA-activated protein kinase (PKR), a serine/threonine kinase, is activated in virus-infected cells and acts as an antiviral machinery of type I interferons. PKR controls several stress response pathways induced by double-stranded RNA, tumor necrosis factor-aor lipo-polysaccharide, which result in the activation of stress-acti-vated protein kinase/c-Jun NH2 -terminal kinase and p38 of the mitogen-activated protein kinase family.

Tumor necrosis factor (TNF)-related apoptosis inducing ligand (TRAIL) has been known to induce tumor-specific apoptosis and to share the structural and functional char-acteristics with the proteins of TNF family. Recently, the crystal structure of humanTRAIL showed that TRAIL is a homotrimeric protein whose subunits contain mainly b-sheets. We characterized the structural changes of recombinant human TRAIL induced byacidification and the biological implication of the structural characteristics at acidic pH in the interaction with the lipid bilayer. ...

Previous results have shown that theoncoembryonicmarker a-fetoprotein (AFP) is able to induce apoptosis in tumor cells through activation of caspase 3, bypassing Fas-dependent and tumor necrosis factor receptor-dependent signaling. In this study we further investigate the molecular interactions involvedin the AFP-mediatedsignaling of apoptosis. We show that AFP treatment of tumor cells is accompaniedby cytosolic translocation of mitochondrial cytochromec.

Mononuclear phagocytes play a pivotal role in the pro-gression of septic shock by producing tumor necrosis factor-a(TNF-a) andother inflammatorymediators in response to lipopolysaccharide (LPS) fromGram-negativebacteria.Our previous studies have shown monocyte and macrophage activationcorrelatewithchanges inmembranephospholipid composition, mediated by acyltransferases. Interferon-c (IFN-c), which activates and primes these cells for enhanced inflammatory responses to LPS, was found to selectively activate lysophosphatidylcholine acyltransferase (LPCAT) (P...

Tumor necrosis factor-a converting enzyme (TACE or ADAM17) is a member of the ADAM (a disintegrin and metalloproteinase) family of type I membrane proteins and mediates the ectodomain shedding of various membrane-anchored signaling and adhesion proteins. TACE is syn-thesized as an inactive zymogen, which is subsequently proteolytically processed to the catalytically active form.We have identified the proprotein-convertases PC7 and furin to be involved in maturation of TACE.

The Goodpasture antigen-binding protein, GPBP, is a serine⁄threonine kinase whose relative expression increases in autoimmune processes. Tumor necrosis factor (TNF) is a pro-inflammatory cytokine implicated in auto-immune pathogenesis. Here we show that COL4A3BP, the gene encoding GPBP, maps head-to-head withPOLK, the gene encoding for DNA polymerase kappa (polj), and shares with it a 140-bp promoter containing a Sp1 site, a TATA-like element, and a nuclear factor kappa B (NFjB)-like site

Exposure of blood to tissue factor (TF) rapidly initiates the coagulation serine protease cascades. TF is expressed by macrophages and other types of cell within atherosclerotic lesions and plays an important role in throm-bus formation after plaque rupture. Macrophage TF expression is induced by pro-inflammatory stimuli including lipopolysaccharide (LPS), inter-leukin-1band tumor necrosis factor-a. Here we demonstrate that activation of liver X receptors (LXRs) LXRaand LXRbsuppresses TF expression....

Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) is amember of theTNFfamilyandapotent inducer of apoptosis. TRAIL has been shown to effectively limit tumor growthin vivowithout detectable cytotoxic side-effects. Interferon (IFN)-coften modulates the anticancer activities of TNF family members including TRAIL. How-ever, little is known about the mechanism. To explore the mechanism, A549, HeLa, LNCaP, Hep3B andHepG2 cells were pretreated with IFN-c, and then exposed to TRAIL.

In order to determine the amino-terminal sequence requirements for protein N-myristoylation, site-directed mutagenesis of the N-terminal region was performed using tumor necrosis factor (TNF) mutants as model substrate proteins. Subsequently, the susceptibility of thesemutants to protein N-myristoylation was evaluated by metabolic labe-ling in anin vitrotranslation systemusing rabbit reticulocyte lysate. A TNF mutant having the sequence MGAAAAA AAA at its N-terminus was used as the starting sequence to identify elements critical for protein N-myristoylation....

The transcription factor nuclear factorjB(NF-jB) is an activator of multiple cytokines, chemokines and adhesion molecules, which are important in inflammatory diseases such as asthma, and is consequently considered as an attractive therapeutic target. In the present study, a con-stitutively active dominant version of IjBa,IjBaDN, was introduced into A549 pulmonary cells by adenovirus-mediated delivery. The dominant IjB, but not a null viral vector, prevented the induction of NF-jB-dependent transcription by both tumor necrosis factora(TNFa)and interleukin-1b(IL-1b). ...

Plasminogen activator inhibitor type 1 (PAI-1) is induced by many proin-flammatory and pro-oxidant factors. Among them, tumor necrosis factora (TNFa), a pivotal early mediator that regulates and amplifies the develop-ment of inflammation, is one of the strongest PAI-1 synthesis activators. Location of the TNFa response element in the PAI-1 promoter is still ambiguous.

Lipopolysaccharide is a major constituent of the outer membrane of Gram-negative bacteria. It activates monocytes and macrophages to produce cytokines such as tumor necrosis factor-aand interleukins IL-1b and IL-6. These cytokines appear to be responsible for the neurotoxicity observed in peripheral nervous system inflammatory disease.
