Vinca alkaloids

Terpenoid indole alkaloids (TIAs) vincristine and vinblastine are two essential anticancer drugs used in cancer treatments. These compounds are naturally available only in Catharanthus roseus in low accumulation, leading to efforts to upregulate TIAs substances through other expression organisms. The combination of vindoline and catharanthine into α-3∼,4∼- anhydrovinblastine (AVLB) is an important step for the biosynthesis of vinca alkaloids, which serves as a precursor to vinblastine and vincristine.

Seventeen strains of endophytic fungi have been isolated from three species of Vinca plants - V. minor, V. erecta and V. rosea collected from different areas of Uzbekistan. These endophytic fungi represented different genera and were screened for vinca alkaloids production. Out of the seventeen endophytic fungal cultures screened, extracts of 5 strains contain compounds of alkaloid nature (vinblastine and vincristine).

Resistance of solid tumors to treatment is significantly attributed to pharmacokinetic reasons at both cellular and multi-cellular levels. Anticancer agent must be bio-available at the site of action in a cytotoxic concentration to exert its proposed activity. P-glycoprotein (P-gp) is a member of the ATP-dependent membrane transport proteins; it is known to pump substrates out of cells in ATP-dependent mechanism. The over-expression of P-gp in tumor cells reduces the intracellular drug concentrations, which decreases the cytotoxicity of a broad spectrum of antitumor drugs.

The taxanes include paclitaxel and docetaxel. These agents differ from the vinca alkaloids in that the taxanes stabilize microtubules against depolymerization. The "stabilized" microtubules function abnormally and are not able to undergo the normal dynamic changes of microtubule structure and function necessary for cell cycle completion. Taxanes are among the most broadly active antineoplastic agents for use in solid tumors, with evidence of activity in ovarian cancer, breast cancer, Kaposi's sarcoma, and lung tumors.
