Brain metastasis is common in patients with non-small cell lung cancer (NSCLC) and it is associated with poorer prognosis. Several options to control the secondary brain tumors in the context include chemotherapy, whole-brain radiation, stereotactic surgery, surgery. However, chemotherapy is ineffective to those patients because of poor penetration through the blood-brain barrier. Whole-brain radiation therapy used to be a standard option for brain metastases.

Mice expressing a C-terminal fragment of the prion protein instead of wild-type prion protein die from massive neuronal degeneration within weeks of birth. The C-terminal region of PrPc (PrP121–231) expressed in these mice has an intrinsic neurotoxicity to cultured neurones. Unlike PrPSc, which is not neurotoxic to neurones lacking PrPc expression, PrP121–231 was more neurotoxic to PrPc-deficient cells. Human mutations E200K and F198S were found to enhance toxicity of PrP121–231 to PrP-knockout neurones and E200K enhanced toxicity to wild-type neurones.

Glycosphingolipids (GSLs) were purified from adults and plerocercoids of the tapeworm Diphyllobothrium hottai, and their chemical structures were determined. Total lipid fractions prepared from chloroform/methanol extracts of whole tissues were fractionated successively on ion-exchange chromatography, silicic acid column chromatography, and preparative TLC. The purified GSLs were characterized by methylation analysis, TLC-immunostaining, liquid secondary ion MS, MALDI-TOF MS, and 1H-NMR.

A library of random peptide sequences was used to select peptides that inhibit an anti-idiotypic catalytic Ig, immuno-globulin (IgG) 9G4H9, withab-lactamase-like activity. This library displays cyclic heptapeptides on the surface of bac-teriophagesand representsacollectionofupto4.5·10 9 peptides. The first selection step aimed at enriching the lib-rary in species that bind to the whole Ig molecule. The sec-ond stepwas todiscriminate peptides that bind topart of the molecule other than the active site. Selected peptides were then screened by surface plasmon resonance analysis.

A cDNA library from tobacco inoculated withRhizoctonia solaniwas con-structed, and several cDNA fragments were identified by differential hybridization screening. One cDNA clone that was dramatically repressed, NtKTI1, was confirmed as a member of the Kunitz plant proteinase inhibi-tor family. RT-PCR analysis revealed that NtKTI1was constitutively expressed throughout the whole plant and preferentially expressed in the roots and stems.
