The consequences of tuberculosis on human society are immense. Tuberculosis remains a major cause of morbidity and mortality in many countries and a significant public health problem worldwide. Active tuberculosis is diagnosed by detecting Mycobacterium tuberculosis complex bacilli in specimens from the respiratory tract (pulmonary TB) or in specimens from other bodily sites (extra pulmonary TB).

The gene fprA of Mycobacterium tuberculosis, encoding a putative protein with 40% identity to mammalian adrenodoxin reductase, was expressed in Escherichia coli and the protein purified to homogeneity. The 50-kDa protein monomer contained one tightly bound FAD, whose fluorescence was fully quenched. FprA showed a low ferric reductase activity, whereas it was very active as a NAD(P)H diaphorase with dyes. Kinetic parameters were determined and the specificity constant (kcat/Km) for NADPH was two orders of magnitude larger than that of NADH....

The Rv0948c gene fromMycobacterium tuberculosisH37Rv encodes a 90 amino acid protein as the natural gene product with chorismate mutase (CM) activity. The protein, 90-MtCM, exhibits Michaelis–Menten kinetics with akcat of 5.5 ± 0.2 s )1 and a Kmof 1500 ± 100lmat 37C and pH 7.5. The 2.0 A˚ X-ray structure shows that 90-MtCM is an alla-helical homodimer (Protein Data Bank ID: 2QBV) with the topology ofEscheri-chia coliCM (EcCM), and that both protomers contribute to each catalytic site.

The gene encompassing ORF Rv1885c with weak sequence similarity to AroQ chorismate mutases (CMs) was cloned from the genome ofMycobac-terium tuberculosis and expressed in Escherichia coli. The gene product (*MtCM) complements a CM-deficient E. colistrain, but only if produced without the predicted N-terminal signal sequence typical ofM. tuberculosis. The mature *MtCM, which was purified by exploiting its resistance to irre-versible thermal denaturation, possesses high CM activity in vitro.

The Mycobacterium tuberculosisgenome contains an unusually high number of proteins involved in the metabo-lism of lipids belonging to the Lip family, including various nonlipolytic and lipolytic hydrolases. Driven by a structural genomic approach, we have biochemically characterized the Rv1399c gene product, LipH, previously annotated as a putative lipase. Rv1399c was overexpressed inE. colias inclusion bodies and refolded. Rv1399c efficiently hydro-lyzes soluble triacylglycerols and vinyl esters.

The genesRv1318c, Rv1319c, Rv1320cand Rv3645 1 of Mycobacterium tuberculosisare predicted to code for four out of 15 adenylyl cyclases in this pathogen. The proteins consist of a membrane anchor, a HAMP region and a class IIIb adenylyl cyclase catalytic domain. Expression and purification of the isolated catalytic domains yielded aden-ylyl cyclase activity for all four recombinant proteins.
