Human cells contain two homologs of the yeast RAD23 protein, hHR23A and hHR23B, which participate in the DNA repair process. hHR23B hou-ses a domain (residues 277–332, called XPCB) that binds specifically and directly to the xeroderma pigmentosum group C protein (XPC) to initiate nucleotide excision repair (NER). This domain shares sequence homology with a heat shock chaperonin-binding motif that is also found in the stress-inducible yeast phosphoprotein STI1.

This work presents a computational analysis of the molecular characteris-tics shared by the adenylation domains from traditional nonribosomal pep-tide synthetases (NRPSs) and the group of the freestanding homologous enzymes: a-aminoadipate semialdehyde dehydrogenase, a-aminoadipate reductase and the protein Ebony. The results of systematic sequence com-parisons allow us to conclude that a specificity-conferring code, similar to that described for the NRPSs, can be recognized in such enzymes.

Molecular phylogeny among catalase–peroxidases, cyto-chromecperoxidases, and ascorbate peroxidases was ana-lysed. Sixty representative sequences covering all known subgroups of class I of the superfamily of bacterial, fungal, and plant heme peroxidases were selected.
