Glycosaminoglycan chain analysis

Domain IV, consisting of about 230 residues, represents a particular protein module so far found only in lamininb1 andb2 chains. Both domains were obtained by recombi-nant production in mammalian cells. They showed a globular structure, as expected from electron microscopic examination of laminins. Fragmentb1IV was obtained as a monomer and a disul®de-bonded dimer, and both were modi®ed to50% by a single chondroitin sulfate chain attached to Ser721 of an SGD consensus sequence.

The importance of proteoglycans for secretionof proteolytic enzymes was studied in the murine macrophage cell line J774. Untreated or 4b-phorbol 12-myristate 13-acetate (PMA)-stimulated macrophages were treated with hexyl-b-D-thioxyloside to interfere with the attachment of glycosaminoglycan chains to their respective protein cores. Activation of the J774 macrophages with PMA resulted in increased secretion of trypsin-like serine proteinase activity.
