BTG/Tob family proteins, which are characterized by similarities in their N-terminal BTG/Tob homology domains, control cell growth negatively. Among the BTG/ Tob family members, BTG2/TIS21/PC3 proteins have been reported to have short lives and to be degraded by the proteasome. However, the mechanisms regulating the stabilities of other BTG/Tob family proteins have not yet been clarified.

A series of chimeric receptors was generated between the epidermal growth factor (EGF) receptor, ErbB-1, and its homologue, ErbB-4, to investigate the roles of the extracellular domains (I–IV) in the ligand specificities. As compared with ErbB-1 and the chimeras with both domains I and III of ErbB-1, the chimeras with only one of these domains exhibited reduced binding of 125I-labeled EGF. Particularly, the contribution of domain III was appreciably larger than that of domain I of ErbB-1 in 125I-labeled EGF binding.

To search for novel regulatory regions, we examined the features of chromatin structure in the rat B29/Ig-b gene and its flanking regions by determining DNase I hypersensitive sites (DHS) in plasmacytoma-derived Y3 cells. Six Y3 cellspecific DHS were detected at )8.6, promoter, +0.7, +4.4, +6.0, and +8.7 kb. The DHS at +4.4, +6.0, and +8.7 kb were present in the intergenic region between B29/Ig-b and growth hormone (GH) genes and were mapped inside conserved sequences in rat and humans.

Recently, we identified novel avian and amphibian hypothalamic neuropeptides that inhibited gonadotropin release and stimulated growth hormone release. They were characterized by a similar structure including theC-terminal LPLRF-NH2motif. To clarify that the expression of these novel hypothalamicneuropeptides is aconservedproperty in vertebrates, we characterized a cDNA encoding a similar novel peptide, having LPLRF-NH2 from the goldfish brain, by a combination of 3¢and 5¢rapid amplification of cDNA ends (RACE). ...

Basic ®broblast growth factor (bFGF) regulates diversi®ed biological functions in rat Sertoli cells. This report demon-strates that bFGF inhibits steroidogenesis in developing rat Sertoli cells. Follicle stimulating hormone (FSH)-stimulated estradiol production was reduced by bFGF. Moreover, the amount of cytochrome P450 aromatase, responsible for the irreversible transformation of androgens into estrogens, is decreased by bFGF at the transcriptional level.

To sustain life mammals have an absolute and continual requirement for oxygen, which is necessary to produce energy for normal cell survival and growth. Hence, main-tainingoxygen homeostasis is a critical requirement and mammals have evolved a wide range of cellular and phy-siological responses to adapt to changes in oxygen avail-ability. In the past few years it has become evident that the transcriptional protein complex hypoxia-inducible factor (HIF) is a key regulator of these processes.

Thyroid hormone has various effects on cell proliferation, growth and apoptosis. To gain more insight into the molecular dynamics caused by thyroid hormone, gene expression in HeLaTRcells that constitutively over-expressed the thyroid hormone receptor (TR) was analyzed. Gene expression profiling of the HeLaTRcells with an oligonucleotide microarray yielded 229 genes whose expression was significantly altered by T3.

accharomyces cerevisiaeis a widely used host in the pro-ductionof therapeutic peptides andproteins.Herewe report the identification of a novel endoprotease in S. cerevisiae. It is encoded by the CYM1gene and is specific for the C-terminus of basic residues of heterologously expressed peptides. Gene disruption ofCYM1not only reduced the intracellular proteolysis, but also enhanced the secretion of heterologously expressed peptides such as growth hormone, pro-B-type natriuretic peptide and pro-cholecystokinin....

Gprotein-coupled receptor 30 (GPR30) has previously been described tobe important in steroid-mediatedgrowthand to inhibit cell proliferation. Here we investigated whether the effect of GPR30 on cell growth is dependent on steroid hormone receptors. We stably introduced GPR30 in immortalizednormalmammaryepithelial (HME) cellsusing retroviruses for gene delivery. GPR30 inhibited the growth and proliferation of the cells. They expressed glucocorticoid receptor, but not estrogen or progesterone receptor....

Among the RFamide peptide groups, PQRFamide peptides, such as neuro-peptide FF (NPFF) and neuropeptide AF (NPAF), share a common C-ter-minal Pro-Gln-Arg-Phe-NH2 motif. LPXRFamide (X¼L or Q) peptides, such as gonadotropin-inhibitory hormone (GnIH), frog growth hormone-releasing peptide (fGRP), goldfish LPXRFamide peptide and mammalian RFamide-related peptides (RFRPs), also share a C-terminal Leu-Pro-Leu⁄Gln-Arg-Phe-NH2 motif.

Primary cultured fibroblasts of four patients with idiopathic short stature and severe growth delay, which displayed normal growth hormone receptor expression presented a reduced ability for activation of signal transducer and activator of transcription-3 (STAT3). Impaired STAT3 activation was accompanied by cell-cycle arrest at the Go⁄G1 phase.

Two H7721 human hepatocarcinoma cell lines showing moderate and high expression of a1,3-fucosyltransferase (FucT)-VII cDNA were established and designated FucTVII-M and FucTVII-H, respectively. In a1,3-FucT-VII-transfected cells, expression of insulin receptor (InR)a- and bsubunits and epidermal growth factor receptor (EGFR) on the cell surface and in cells, as well as the sialyl Lewis X (SLe x , the product of a1,3-FucT-VII) content of the EGFR were unchanged.

The crustacean hyperglycemic hormone (CHH) plays an important role in the regulation of hemolymph glucose levels, but it is also involved in other functions such as growth, molting and reproduction. In the present study we describe the first CHH family gene isolated from the Atlantic Ocean shrimp Litopenaeus schmitti.

O-Fucosylation is a post-translational glycosylation in which anO-fucose is covalently attached to the hydroxyl group of a specific serine or threon-ine residue. This modification occurs within the consensus sequence C2X4)5(S⁄T)C3 present on epidermal growth factor-like repeats of several proteins, including the Notch receptors and their ligands.

Peptides function as chemical signals between cells of multicellular organ-isms via specific receptors on target cells. Many hormones, neuromodula-tors and growth factors are peptides. Peptide hormones and other biologically active peptides are synthesized as higher molecular weight pre-cursor proteins (pro-hormones), which must undergo post-translational modification to yield the bioactive peptide(s).

Ghrelin, the endogenous ligand for the growth hormone secretagogue receptor type 1a (GHS-R1a), is a 28 amino acid residue with a post-trans-lational octanoyl modification on Ser3. Despite the biomedical interest in this hormone, the fine details of its regulation and the mechanisms control-ling its secretion are largely unknown.
