Hepatitis A

Hepatitis B infection remains a global health problem, with progression to acute-chronic hepatitis, severe liver failure, and death making hepatitis B one of the most serious infections worldwide. The disease is most widely transmitted from an infected mother to her baby, after exposure to infected blood or body fluids or unsafe sexual contact.

Polymerase Chain Reaction is widely held as one of the most important inventions of the 20th century in molecular biology. Small amounts of the genetic material can now be amplified to be able to a identify, manipulate DNA, detect infectious organisms, including the viruses that cause AIDS, hepatitis, tuberculosis, detect genetic variations, including mutations, in human genes and numerous other tasks.

The current research project genotyped HAV RNA by using protocols and primer sets designed by HAVNET. It collected and genotyped previous HAV-positive samples in VIDRL sample bank, and the results were used to establish epidemiological and molecular database of the HAV strains. The database was used to investigate the relationship between the current diagnostic samples and the previous HAV infection cases. It was expected to identify the source of the hepatitis A infection.

Hepatitis B Infection is an occupational high risk factor. Immune response for vaccination was measured by Antibody to Hepatitis B surface antigen (anti HBs) levels. A course of Hep B vaccine yields a protective immune response (anti-HBs ≥10 mIU/mL) this study was designed to determine the anti HBs levels in Health Care Workers (HCWs) working as laboratory technician and staff nurses.

Schistosomiasis is an endemic disease in Egypt caused by the trematode Schistosoma which has different species. c the best known form of chronic disease with a wide range of clinical manifestations. The pathogenesis of schistosomiasis is related to the host cellular immune response. This leads to granuloma formation and neo angiogenesis with subsequent periportal fibrosis manifested as portal hypertension, splenomegaly and esophageal varices. Intestinal schistosomiasis is another well identified form of chronic schistosomal affection.

Wilson disease (WD), an inherited disorder affecting copper metabolism, is characterized by hepatic cirrhosis and neuronal degeneration, which result from toxic levels of copper that accumulate in the liver and brain, respectively. We reported previously that the 1.3-kb promoter of the WD gene contains four metal response elements (MREs). Among the four MREs, MREa plays the most important role in the transcriptional activation of the WD promoter.

Glycolysis from [6-3 H]glucose and gluconeogenesis from [U-14 C]glycerol were examined in isolated hepatocytes from fasted rats. A 5 mMbolus of glycerol inhibited phosphory-lationof 40 mMglucoseby50%andglycolysis bymore than 60%, and caused cellular ATP depletion and glycerol 3-phosphate accumulation. Gluconeogenesis from 5 mM glycerol was unaected by the presence of 40 mMglucose.

Fructose 2,6-bisphosphate (Fru-2,6-P2 ) plays an important role in the regulation of major carbohydrate fluxes as both allosteric activator and inhibitor of target enzymes. To examine the role of Fru-2,6-P2in the regulation of hepatic carbohydrate metabolism in vivo,Fru-2,6-P2 levels were elevated in ADM mice with adenovirus-mediated overex-pression of a double mutant bifunctional enzyme, 6-phos-phofructo-2-kinase/fructose-2,6-bisphosphatase (n¼6), in comparison to normal control mice (control,n¼6). ...

Hexadecylphosphocholine (HePC) is a synthetic lipid rep-resentative of a new group of antiproliferative agents, alkylphosphocholines (APC), which are promising candi-dates in anticancer therapy. Thus we have studied the action of HePC on the human hepatoblastoma cell line HepG2, which is frequently used as a model for studies into hepatic lipid metabolism.

Hepatopoietin (HPO) is a novel human hepatotrophic growth factor. Recently, we demonstrated that the extra-cellular ligand form of HPO can stimulate proliferation of hepatic cells via its specific receptor, which is on the surface of these cells. Also, the intracellular formofHPOpotentiates the transcriptional factor AP-1. Intriguingly, a variety of HPO complexes with different molecular masses were detected in hepatocytes.

Although efficient vaccines are available, chronic hepatitis B (HBV) infection poses a major health problem worldwide, and prolonged treatment of chronicallyinfected HBV patients with nucleoside analogs often results in drug-resistantHBVvariants.Therefore, it is critical toevaluate the contribution of the HBV polymerase to mutations. FLAG-taggedwild-type (FPolE) andmutant (FPolE/D551A)HBV polymerases have been expressed in insect cells and purified. The purified FPolE showed DNA polymerase activity, but FPolE/D551A did not, implying that the activity was derived from FPolE....

Multiple signalling pathways are involved in themechanism by which insulin stimulates hepatic glycogen synthesis. In this study we used selective inhibitors of glycogen synthase kinase-3 (GSK-3) and an allosteric inhibitor of phosphory-lase (CP-91149) that causes dephosphorylation of phos-phorylasea, to determine the relative contributions of inactivation of GSK-3 and dephosphorylation of phos-phorylaseaas alternative pathways in the stimulation of glycogen synthesis by insulin in hepatocytes.

The immune coagulant fgl2/fibroleukin has been previously shown to play a pivotal role in the pathogenesis of murine and human fulminant hepatitis and fetal loss syndrome. Constitutive expression of fgl2 transcripts at low levels are seen in cytotoxic T cells, endothelial, intestinal and tropho-blast cells, while specific factors (such as virus and cytokines) are required to induce high levels of fgl2 expression in other cell types including monocytes/macrophages.

A search has been initiated for lead inhibitors of the non-structural protein 3 (NS3)-associated NTPase/helicase activities of hepatitis C virus, the related West Nile virus, Japanese encephalitis virus and the human mitochondrial Suv3 enzyme. Random screening of a broad range of unre-lated low-molecular mass compounds, employing both RNA and DNA substrates, revealed that 4,5,6,7-tetra-bromobenzotriazole (TBBT) hitherto known as a potent highly selective inhibitor of protein kinase 2,is a good inhibitor of the helicase, but not NTPase, activity of hepa-titis C virus NTPase/helicase. ...

The humanSUV3gene encodes an NTP-dependent DNA⁄RNA DExH box helicase predominantly localized in mitochondria. Its orthologue in yeast is a component of the mitochondrial degradosome complex involved in the mtRNA decay pathway. In contrast to this, the physiological func-tion of human SUV3remains to be elucidated. In this report we demon-strate that the hSuv3 protein interacts with HBXIP, previously identified as a cofactor of survivin in suppression of apoptosis and as a protein that binds the HBx protein encoded by the hepatitis B virus....

We have defined thein vivo andin vitro metabolic fate of internalized chol-era toxin (CT) in the endosomal apparatus of rat liver. In vivo, CT was internalized and accumulated in endosomes where it underwent degrada-tion in a pH-dependent manner. In vitro proteolysis of CT using an endo-somal lysate required an acidic pH and was sensitive to pepstatin A, an inhibitor of aspartic acid proteases.
