In this work, the kinetics rates have been measured by optical microscopy using a single particle approach. The advantages of this analytical technique are an easier management of species transport in solution and a precise following of the dissolution rate.

Study of high temperature steam oxidation kinetics during the high temperature oxidation was carried out on the sponge base E110 cladding tubes material in the temperature range 600–1300 °C. The oxidation kinetics derived from the weight gain measurements showed a parabolic rate law for temperatures 1100 °C and higher only.

Hepcidin (H25) is a hormone peptide synthesized by the liver that binds to ferroportin and blocks iron export. In this study, H25 was inhibited by administration of single and multiple doses of an anti-H25 monoclonal antibody Ab 12B9m in cynomolgus monkeys. The objective of this analysis was to develop a pharmacodynamic model describing the role of H25 in regulating iron homeostasis and the impact of hepcidin inhibition by Ab 12B9m. Total serum H25 and Ab 12B9m were determined in each animal. Corresponding measurements of serum iron and hemoglobin (Hb) were obtained.

Nanostructured Au-Ceria has been known as a promising catalyst for the low-temperature water-gas shisft reaction. The catalyst prepared by coprecipitation method with the gold loading various between 2 - 3 at.% with the crystallite size of 2.9 nm has been used to study some factors that effect to catalytic activity and long-term stability of this material.

Electrodeposition kinetics of Co thin film were investigated and magnetic coercivity of the deposited films were studied using potentiostatic technique and magnetic hyteresis loop measurements. Results showed that the nucleation mechanism of the electrodeposition process changed from instantaneous mode to progressive mode when potential was changed from –0.7 V to –0.8 V. The magnetic coercivity Hc increases with deposition time and the film with Hc < 15 Oe can be obtained with deposition time t < 5 s.

Pyruvate decarboxylase (EC 4.1.1.1) was isolated and purified from the yeast Kluyveromyces lactis. The properties of this enzyme relating to the native oligomeric state, the subunit size, the nucleotide sequence of the coding gene(s), the catalytic activity, and protein fluorescence as well as circular dichroism are very similar to those of the well characterized pyruvate decarboxylase species from yeast.

VanXYC, a bifunctional enzyme from VanC-phenotype Enterococcus gallinarum BM4174 that catalyses D,D-peptidase and D,D-carboxypeptidase activities, was purified as the native protein, as a maltose-binding protein fusion and with an N-terminal tag containing six histidine residues. The kinetic parameters of His6–VanXYC were measured for a variety of precursors of peptidoglycan synthesis involved in resistance: for D-Ala-D-Ala, the Km was 3.6 mM and kcat, 2.5 s)1; for UDP-MurNAc-L-Ala-D-Glu-L-Lys-D-Ala-DAla (UDP-MurNAc-pentapeptide[Ala]), Km was 18.8 mM and kcat 6.

Cloning and over-expression of human glucose 6-phosphate dehydrogenase (Glc6P dehydrogenase) has for the first time allowed a detailed kinetic study of a preparation that is genetically homogeneous and in which all the protein molecules are of identical age. The steady-state kinetics of the recombinant enzyme, studied by fluorimetric initial-rate measurements, gave converging linear Lineweaver–Burk plots as expected for a ternary-complex mechanism.

The kinetics and thermodynamics of the urea-induced unfolding of ¯avodoxin and apo¯avodoxin fromDesulfo-vibrio vulgariswere investigated by measuring changes in ¯avin and protein ¯uorescence. The reaction of urea with ¯avodoxin is up to 5000 times slower than the reaction with the apoprotein (0.67 s )1 in 3 Murea in 25 mMsodium phosphate at 25°C), and it results in the dissociation of FMN.

Báo cáo Y học: Fluorescence study of the high pressure-induced denaturation of skeletal muscle actin
Ikkai&Ooi [Ikkai,T.&Ooi,T. (1966)Biochemistry5, 1551± 1560] made a thorough study of the eect of pressure on G- and F-actins. However, all of the measurements in their study were made after the release of pressure. In the present experiment in situ observations were attempted by using eATP to obtain further detailed kinetic and thermodynamic information about the behaviour of actin under pressure.

The mechanism by which the fatty acid (1,4)-desaturase of Calendulaofficinalisproduces calendicacid fromlinoleicacid has been probed through the use of kinetic isotope effect (KIE) measurements. This was accomplished by incubating appropriate mixtures of linoleate and regiospecifically dideuterated isotopomers with a strain ofSaccharomyces cerevisiaeexpressing a functional (1,4)-desaturase.

Liver microsomal preparations are routinely used to predict drug interactions that can occurin vivo as a result of inhi-bition of cytochrome P450 (CYP)-mediated metabolism. However, the concentration of free drug (substrate and inhibitor)at its intrahepatic site of action, a variable that cannot be directly measured, may be significantly different from that in microsomal incubation systems.

The process of pressure-induced modification of horse liver alcohol dehydrogenase (HLADH) was followed by measuringin situ catalytic activity (up to 250 MPa), intrinsic fluorescence (0.1–600 MPa) and modifications of FTIR spectra (up to 1000 MPa). The tryptophan fluor-escence measurements and the kinetic data indicated that the pressure-induced denaturation of HLADH was a process involving several transitions and that the observed transient states have characteristic properties of molten globules. ...

To better define the mechanism of membrane protein insertion into the membrane of the endoplasmic reticulum, we measured the kinetics of translocation across microsomal membranes of the N-terminal lumenal tail and the lumenal domain following the second transmembrane segment (TM2) in the multispanning mouse protein Cig30. In the wild-type protein, the N-terminal tail translocates across the membrane before the down-stream lumenal domain.

The oligomeric state and kinetics of ligand binding were measured for wild-type cytoglobin. Cytoglobin has the classical globin fold, with an extension at each extremity of about 20 residues. The extended length of cytoglobin leads to an ambiguous interpretation of its oligomeric state.

Realistic quantitative models require data from many laboratories. There-fore, standardization of experimental systems and assay conditions is crucial. Moreover, standards should be representative of thein vivoconditions. How-ever, most often, enzyme–kinetic parameters are measured under assay con-ditions that yield the maximum activity of each enzyme.
