One-electron oxidation of six different c-type lysozymes from hen egg white, turkey egg white, human milk, horse milk, camel stomach and tortoise was studied by gamma-and pulse-radiolysis.In the first step, one tryptophan side chain is oxidized to indolyl free radical, which is produced quantitatively.As shown already, the indolyl radical subse-quently oxidizes a tyrosine side chain to the phenoxy radical in an intramolecular reaction.However this reaction is not total and its stoichiometry depends on the protein.Rate constants also vary between proteins, from 120Æs )1 to 1000Æs )1 at pH 7.

Proteins tend to form inactive aggregates at high tempera-tures. We show that polyamines, which have a relatively simple structure as oligoamids, effectively prevent thermal inactivation and aggregation of hen egg lysozyme. In the presence of additives, including arginine and guanidine (100 mM), more than 30% of 0.2 mgÆmL )1 lysozyme in sodium phosphate buffer (pH 6.5) formed insoluble aggre-gates by heat treatment (98C for 30 min). 1

Arginine is a versatile additive to prevent protein aggrega-tion. This paper shows that arginine ethylester (ArgEE) prevents heat-induced inactivation and aggregation of hen egg lysozyme more effectively than arginine or guanidine. TheadditionofArgEEdecreased themelting temperatureof lysozyme. This data could be interpreted in terms of ArgEE binding to unfolded lysozyme, possibly through the ethyl-ated carboxyl group, which leads to effective prevention of intermolecular interaction among aggregation-prone mole-cules.

We report the secreted expression byPichia pastorisof two human lyso-zyme variants F57I and W64R, associated with systemic amyloid disease, and describe their characterization by biophysical methods. Both variants have a substantially decreased thermostability compared with wild-type human lysozyme, a finding that suggests an explanation for their increased propensity to form fibrillar aggregates and generate disease.

We have reported previously that the calcium-depleted form of bovine a-lactalbumin (apoa-LA) interacts with hen egg-white lysozyme (LYS) to form spherical supramolecular structures. These supramolecular structures contain an equimolar ratio of the two proteins. We further explore here the organization of these structures.

Protein oligomeric complexes have emerged as a major target of current research because of their key role in aggregation processes in living systems andin vitro. Hydrophobic and charged surfaces may favour the self-assembly process by recruiting proteins and modifying their interactions. We found that equine lysozyme assembles into multimeric complexes with oleic acid (ELOA) at the solid–liquid interface within an ion-exchange chromatography column preconditioned with oleic acid.

In ruminants, some leaf-eating animals, and some insects, defensive lyso-zymes have been adapted to become digestive enzymes, in order to digest bacteria in the stomach. Digestive lysozyme has been reported to be resis-tant to protease and to have optimal activity at acidic pH. The structural basis of the adaptation providing persistence of lytic activity under severe gastric conditions remains unclear.

Humana-lactalbumin made lethal to tumor cells (HAMLET) and equine lysozyme with oleic acid (ELOA) are complexes consisting of protein and fatty acid that exhibit cytotoxic activities, drastically differing from the activity of their respective proteinaceous compounds. Since the discovery of HAMLET in the 1990s, a wealth of information has been accumulated, illuminating the structural, functional and therapeutic properties of protein complexes with oleic acid, which is summarized in this review.
