Human amylin is a small fibrillogenic protein that is the major constituent of pancreatic islet amyloid, which occurs in most subjects with type 2 dia-betes. The crystal structure of human pancreatic cationic trypsin showed the chemical modification of Tyr154, which was originally described as phos-phorylation [Gaboriaud C, Serre L, Guy-Crotte O, Forest E & Fontecilla-Camps JC (1996)J Mol Biol259, 995–1010]. Here we report that Tyr154 is sulfated, not phosphorylated.

Chromosomal rearrangements apparently account for the presence of a pri-mate-specific gene (protease serine 3) in chromosome 9. This gene encodes, as the result of alternative splicing, both mesotrypsinogen and trypsino-gen 4. Whereas mesotrypsinogen is known to be a pancreatic protease, neither the chemical nature nor biological function of trypsinogen 4 has been explored previously.

The ATP-sensitive potassium (KATP) channel couples glucose metabolism to insulin secretion in pancreatic b-cells. It comprises regulatory sulfonyl-urea receptor 1 and pore-forming Kir6.2 subunits. Binding and⁄or hydro-lysis of Mg-nucleotides at the nucleotide-binding domains of sulfonylurea receptor 1 stimulates channel opening and leads to membrane hyperpolari-zation and inhibition of insulin secretion.

Expression of the tumor suppressor p16 INK4a after stable transfection can restore the susceptibility of epithelial tumor cells to anoikis. This property is linked to increases in the expression and cell-surface presence of the fibronectin receptor. Considering its glycan chains as pivotal signals, we assumed an effect of p16 INK4a on glycosylation.

The tumor suppressor p16 INK4a has functions beyond cell-cycle control via cyclin-dependent kinases. A coordinated remodeling ofN- and O-glycosyla-tion, and an increase in the presentation of the endogenous lectin galectin-1 sensing these changes on the surface of p16 INK4a -expressing pancreatic carcinoma cells (Capan-1), lead to potent pro-anoikis signals.

Onconase (ONC) is an amphibian member of the pancreatic ribonuclease superfamily that is selectively toxic to tumor cells. It is a much less efficient enzyme than the archetypal ribonuclease A and, in an attempt to gain fur-ther insight, we report the first atomic resolution crystal structure of ONC, determined in complex with sulfate ions at 100 K.
