Shikimate kinase was chosen as a convenient representative example of the subclass of a/b proteins with which to examine the mechanism of protein folding. In this paper we report on the refolding of the enzyme after denaturation in urea. As shown by the changes in secondary and tertiary structure monitored by far UV circular dichroism (CD) and fluorescence, respectively, the enzyme was fully unfolded in 4 M urea. From an analysis of the unfolding curve in terms of the two-state model, the stability of the folded state could be estimated as 17 kJÆmol)1.

Certain amino acids, like glutamine and leucine, induce an anabolic response in liver. They activate p70 ribosomal protein S6 kinase (p70S6K) and acetyl-CoA carboxylase (ACC) involved in protein and fatty acids synthesis, respectively. In contrast, the AMP-activated protein kinase (AMPK), which senses the energy state of the cell and becomes activated under metabolic stress, inactivates by phosphorylation key enzymes in biosynthetic pathways thereby conserving ATP.

Penaeidins are a family of antimicrobial peptides constitutively produced and stored in the haemocytes of penaeid shrimp. In response to microbial stimulation, they are released into the blood circulation and they further attach to shrimp cuticle surfaces through a chitin-binding property. In the present paper, we have analysed their expression, regulation and distribution in shrimp tissues in response to experimental microbial challenge.

Glyoxal is a key compound involved in glyoxal oxidase (GLOX)-dependent production of glyoxylate, oxalate and H2O2 by lignin-degrading basidiomycetes. In this paper, we report that glyoxal was produced from a metabolite of ligninolytic fungi, linoleic acid, by manganese peroxidase (MnP)-dependent lipid peroxidation. In the absence of the parent substrate of linoleic acid, the dialdehyde was oxidized by MnP and Mn(III) chelate to start free radical reactions with emission of chemiluminescence at 700– 710 nm. ...

A cytosolic thyroid-hormone-binding protein (xCTBP), predominantly responsible for the major binding activity of T3 in the cytosol of Xenopus liver, has been shown to be identical to aldehyde dehydrogenase class 1 (ALDH1) [Yamauchi, K., Nakajima, J., Hayashi, H., Horiuchi, R. & Tata, J.R. (1999) J. Biol. Chem. 274, 8460–8469]. Within this paper we surveyed which signaling, and other, compounds affect the thyroid hormone binding activity and aldehyde dehydrogenase activity of recombinant Xenopus ALDH1 (xCTBP/xALDH1) while examining the relationship between these two activities....

In this paper we address the problem of extracting features relevant for predicting protein±protein interaction sites from the three-dimensional structures of protein complexes. Our approach is based on information about evolutionary con-servation and surface disposition. We implement a neural network based system, which uses a cross validation proce-dure and allows the correct detection of 73% of the residues involved in protein interactions in a selected database comprising 226 heterodimers.

This paper describes violaxanthin de-epoxidation in model lipid bilayers. Unilamellar egg yolk phosphatidylcholine (PtdCho) vesicles supplemented with monogalactosyldi-acylglycerol were found to be a suitable system for studying this reaction. Such a system resembles more the native thylakoid membrane and offers better possibilities for studying kinetics and factors controlling de-epoxidation of violaxanthin than a systemcomposed only of monogalacto-syldiacylglycerol and is commonly used in xanthophyll cycle studies. ...

This paper deals with the purification and the partial char-acterization of glutathione S-transferase (GST) isoforms from the clam Ruditapes decussatus. For the first step of purification, two affinity columns, reduced glutathione (GSH)–agarose and S-hexyl GSH–agarose, were mounted in series. Four affinity fractionswere thus recovered. Further purification was performed using anion exchange chroma-tography. Seven fractions, whichpresent aGSTactivitywith 1-chloro-2,4-dinitrobenzene (CDNB) as substrate, were collected and analyzed by RP-HPLC....

A new approach of predicting structural classes of protein domain sequences is presented in this paper. Besides the amino acid composition, the composition of several dipeptides, tripeptides, tetrapeptides, pentapeptides and hexapeptides are taken into account based on the step-wise discriminant analysis. The result of jackknife test shows that this new approach can lead to higher pre-dictive sensitivityand specificityfor reduced sequence similaritydatasets.

Achromobacterprotease I (API) has a unique region of aromatic ring stacking with Trp169–His210in close proxi-mity to the catalytic triad. This paper reveals the electrostatic role of aromatic stacking in the shift in optimum pH to the alkaline region, which is the highest pH range (8.5–10) among chymotrypsin-type serine proteases. The pH-activity profile of API showed a sigmoidal distribution that appears at pH 8–10, with a shoulder at pH 6–8.

The serotype a-specific polysaccharide antigen ofActinoba-cillus actinomycetemcomitansis an unusual sugar, 6-deoxy-D-talose.Guanosinediphosphate (GDP)-6-deoxy-D-talose is the activated sugar nucleotide form of 6-deoxy-D-talose, which has been identified as a constituent of only a few microbial polysaccharides. In this paper, we identify two genes encoding GDP-6-deoxy-D-talose synthetic enzymes, GDP-a-D-mannose 4,6-dehydratase and GDP-4-keto-6-deoxy-D-mannose reductase, in the gene cluster required for the biosynthesis of serotype a-specific polysaccharide anti-gen from A.
