The structure of the new acid and data of potentiometric titration were analyzed in detail before calculating and assigning the pKa values to suitable functional groups. These results will be applied to our further studies of this compound.

The present study investigates teachers‘ perceptions of the values of the CEFR, the perceived readiness and necessity of its application, and the work involved in its application process. Also, it explores teachers‘ responses to the use of the CEFR to renew the general English curriculum, reflected in how they changed their teaching activities, adapted the assigned textbooks and modified their assessment practice. The study was a case study applying the mixed method sequential explanatory model (Creswell & Clark, 2007).

Data abstraction refers to, providing only essential information to the outside word and hiding their background details ie. to represent the needed information in program without presenting the details. Data abstraction is a programming (and design) technique that relies on the separation of interface and implementation.

A clear difference in the enthalpy changes derived from spectroscopic and calorimetric measurements has recently been shown. The exact interpretation of this deviation varied from study to study, but it was generally attributed to the non-two-state transition and heat capacity change. Although the temperature-dependent thermodynamics of the duplex formation was often implied, systemic and extensive studies have been lacking in universally assigning the appropriate thermodynamic parameter sets.

Strictosidine glucosidase (SG) is an enzyme that catalyses the second step in the biosynthesis of various classes of monoterpenoid indole alkaloids. Based on the comparison of cDNA sequences of SG from Catharanthus roseus and raucaffricine glucosidase (RG) from Rauvolfia serpentina, primers for RT-PCR were designed and the cDNA encoding SG was cloned from R. serpentina cell suspension cultures. The active enzyme was expressed in Escherichia coli and purified to homogeneity. Analysis of its deduced amino-acid sequence assigned the SG from R. serpentina to family 1 of glycosyl hydrolases.

Short-chaindehydrogenases/reductases (SDRs) are enzymes of great functional diversity. Even at sequence identities of typically only 15–30%, specific sequence motifs are detect-able, reflecting common folding patterns. We have devel-oped a functional assignment scheme based on these motifs and we find five families. Two of these families were known previously and are calledclassical andextended families, but they are now distinguished at a further level based on coenzyme specificities.

Two-dimensional NMR and molecular dynamics simula-tions have been used to determine the three-dimensional structures of two hairpin DNA structures: d-CTAGAG GATCCUTTTGGATCCT (abbreviated as U1-hairpin) and d-CTAGAGGATCCTTUTGGATCCT (abbreviated as U3-hairpin). The 1 H resonances of both of these hairpin structures have been assigned almost completely.

In the case of oxidized plastocyanin fromSynechocystissp. PCC6803, an NMR approach based on classical two and three dimensional experiments for sequential assignment leaves unobserved 14 out of 98 amino acids. A protocol which simply makes use of tailored versions of 2D HSQC and 3D CBCA(CO)NH and CBCANH leads to the identi-fication of nine of the above 14 residues.

Pest insects such asHelicoverpaspp. frequently feed on plants expressing protease inhibitors. Apparently, their digestive system can adapt to the presence of protease inhibitors. To study this, a trypsin enzymewas purified from the gut of insects that were raised on an inhibitor-containing diet. The amino-acid sequence of this enzyme was analysed by tandem MS, which allowed assignment of 66% of the mature protein amino acid sequence. This trypsin, called HzTrypsin-S, corresponded to a known cDNA sequence fromHelicoverpa. ...

Scorpion toxins that affect sodium channel (NaCh) gating in excitable cells are divided into a-andb-classes. Whereas a-toxinshavebeen found inscorpions throughout theworld, anti-mammalianb-toxins have been assigned, thus far, to New World scorpions while anti-insect selective b-toxins (depressant and excitatory) have been described only in the Old World. 2 This distribution suggested that diversification ofb-toxins into distinct pharmacological groups occurred after the separation of the continents, 150 million years ago....

Ech hydrogenase fromMethanosarcina barkeriis a member of a distinct group of membrane-bound [NiFe] hydrogenases with sequence similarity to energy-conserving NADH:quinone oxidoreductase (complex I). The sequence of the enzyme predicts the binding of three [4Fe-4S] clusters, one by subunit EchC and two by subunit EchF. Previous studies had shown that two of these clusters could be fully reduced under 10 5 Pa of H2at pH 7 giving rise to two distinct S½ electron paramagnetic resonance (EPR) signals, designated as the g¼1.89 and theg¼1.92 signal....

The cellular retinoic acid-binding protein type I (CRABPI) is encoded by a single gene in mammals. We have characterized twocrabp1genes in zebra-fish, designatedcrabp1a and crabp1b. These twocrabp1 genes share the same gene structure as the mammalianCRABP1genes and encode proteins that show the highest amino acid sequence identity to mammalian CRAB-PIs. The zebrafishcrabp1aandcrabp1bwere assigned to linkage groups 25 and 7, respectively.

The AAA-peroxins Pex1p and Pex6p play a critical role in peroxisome bio-genesis but their precise function remains to be established. These two peroxins consist of three distinct regions (N, D1, D2), two of which (D1, D2) contain a conserved230 amino acid cassette, which is common to all ATPases associated with various cellular activities (AAA). Here we show that Pex1p and Pex6p from Saccharomyces cerevisiaedo interact in vivo. We assigned their corresponding binding sites and elucidated the importance of ATP-binding and -hydrolysis of Pex1p and Pex6p for their interaction. ...

In the postgenomic era it is essential that protein sequences are annotated correctly in order to help in the assignment of their putative functions. Over 1300 proteins in current pro-tein sequence databases are predicted to contain a PAS domainbaseduponaminoacid sequence alignments.Oneof theproblemswith the current annotationof thePASdomain is that this domain exhibits limited similarity at the amino acid sequence level.

We have isolated a nonfucosylated and three variously fucosylated neutral oligosaccharides from human milk that are based on theiso-lacto-N-octaose core. Their structures were characterized by the combined use of electrospray mass spectrometry (ES-MS) and NMR spectroscopy. The branching pattern and blood group-related Lewis deter-minants, together with partial sequences and linkages of these oligosaccharides, were initially elucidated by high-sensitivity ES-MS/MS analysis, and then their full structure assignment was completed by methylation analysis and 1 H-NMR.
