Bayesian kernel machine regression (BKMR) analysis was used to evaluate the univariate contaminant exposure effect as well as the contaminant mixture effects on levels of thyroid hormones. Significant and positive associations were found between total T3 and PC-2 (high positive nickel and cadmium loadings), total T3 and PC-3 (negative association with negative loading for nickel and positive loading for cadmium) and TSH and PC-1 (high positive loadings for organic contaminants).

Activating mutations of the thyroid-stimulating hormone receptor (TSHr) have been identified as a cause of toxic adenomas. Germline-inactivating TSHr mutations have beendescribedas a cause of congenital hypothyroidism. The effects of combining activating and inactivating mutations within a single receptor was studied. The double mutant T477I/P639S contained an activating TSHr mutation (P639S) togetherwithan inactivatingone (T477I). Theother one (I486M/P639S) contained two activating mutations.

Thyroid hormone has various effects on cell proliferation, growth and apoptosis. To gain more insight into the molecular dynamics caused by thyroid hormone, gene expression in HeLaTRcells that constitutively over-expressed the thyroid hormone receptor (TR) was analyzed. Gene expression profiling of the HeLaTRcells with an oligonucleotide microarray yielded 229 genes whose expression was significantly altered by T3.

By employing RT-PCR in conjunction with 3¢-RACE, a full-length cDNA encoding a novel zebrafish cytosolic sulfotransferase (SULT) was cloned and sequenced. Sequence analysis revealed that this zebrafish SULT (desig-nated SULT1 ST5) is, at the amino acid sequence level, close to 50% identical to human and dog SULT1B1 (thyroid hormone SULT). A recombinant form of zebrafish SULT1 ST5 was expressed using the pGEX-2TK bacterial expression system and purified from transformed BL21 (DE3) cells.

The relationship between the structure of the N-terminal sequence of trans-thyretin (TTR) and the binding of thyroid hormone was studied. Transthyretin is a tetrameric binding protein involved in the transport of thyroid hormones and in the cotransport of retinol by forming a complex in plasma with retinol-binding protein. In the present study, we report the crystal structure of a macromolecular complex, in which human transthy-retin, human holo-retinol-binding protein and a murine anti-retinol-binding protein Fab are assembled according to a 1 : 2 : 2 stoichiometry.

The structure of the thyroid hormone distributor protein, transthyretin, has been highly conserved during the evolution of vertebrates. Over the last decade, studies into the evolution of transthyretin have revealed the exis-tence of a transthyretin homolog, transthyretin-like protein, in all king-doms. Phylogenetic studies have suggested that the transthyretin gene in fact arose as a result of a duplication of the transthyretin-like protein gene in early protochordate evolution.

A survey of the expression of the transthyretin and thyroxine-binding glob-ulin genes in various species during development provides clues as to how the present thyroid hormone distribution network in extracellular compart-ments developed during vertebrate evolution. Albumin may be the ‘oldest’ component of the thyroid hormone distribution network as it is found in the plasma of all vertebrates investigated.

During vertebrate evolution, the N-terminal region of transthyretin (TTR) subunit has undergone a change in both length and hydropathy. This was previously shown to change the binding affinity for thyroid hormones (THs). However, it was not known whether this change affects other functions of TTR.
