Nội dung nghiên cứu của luận án là khảo sát ảnh hưởng của LXN và RMK đến các enzyme chuyển hóa carbohydrate gồm các enzyme là glucose-6-phosphatase (G6Pase), glucose-6- phosphate dehydrogenase (G6PDH) và lactate dehydrogenase (LDH) in vitro. Khảo sát khả năng chống stress oxy hóa và bảo vệ tế bào tụy tạng min6 khỏi sự chết do tunicamycin gây ra của LXN và RMK.

Porcine heart cytoplasmic malate dehydrogenase (s-MDH) is a dimeric protein (2 · 35 kDa). We have studied equilibrium unfolding and refolding of s-MDH using activity assay, fluorescence, far-UV and near-UV circular dichroism (CD) spectroscopy, hydrophobic probe-1-anilino8-napthalene sulfonic acid binding, dynamic light scattering, and chromatographic (HPLC) techniques. The unfolding and refolding transitions are reversible and show the presence of two equilibrium intermediate states.

Recent years have witnessed high levels of activity in identifying enzyme systems that catalyse H-transfer by quantum tunneling. Rather than being restricted to a small number of specific enzymes as perceived initially, it has now become an accepted mechanism for H-transfer in a growing number of enzymes. Furthermore, H-tunneling is driven by the thermally induced dynamics of the enzyme. In some of those enzymes that break stable C–H bonds the reaction proceeds purely by quantum tunneling, without the need to partially ascend the barrier.

To examine functions of two small heat shock proteins of Escherichia coli, IbpA and IbpB, we constructed His–IbpA and His–IbpB, in which a polyhistidine tag was fused to the N-terminals. Both purified His–IbpA and His–IbpB formed multimers, which have molecular masses of about 2.0– 3.0 MDa and consist of about 100–150 subunits.

The involvement of the lysine residue present at the active site of Ehrlich ascites carcinoma (EAC) cell glyceraldehyde-3-phosphate dehydrogenase (Gra3P DH) was investigated by using the lysine specific reagents trinitrobenzenesulfonic acid (TNBS) and pyridoxal phosphate (PP). Both TNBS and PP inactivated EAC cell Gra3P DH with pseudo-first-order kinetics with the rate dependent on modifier concentration. Kinetic analysis, including a Tsou plot, indicated that both TNBS and PP apparently react with one lysine residue per enzyme molecule.

We previously reported that GTS1 is involved in regulating ultradian oscillations of the glycolytic pathway induced by cyanide in cell suspensions as well as oscillations of energy metabolism in aerobic continuous cultures. Here, we screened a yeast cDNA library for proteins that bind to Gts1p using the yeast two-hybrid system and cloned multiple TDH cDNAs encoding the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase (GAPDH).

Cloning and over-expression of human glucose 6-phosphate dehydrogenase (Glc6P dehydrogenase) has for the first time allowed a detailed kinetic study of a preparation that is genetically homogeneous and in which all the protein molecules are of identical age. The steady-state kinetics of the recombinant enzyme, studied by fluorimetric initial-rate measurements, gave converging linear Lineweaver–Burk plots as expected for a ternary-complex mechanism.

A cytosolic thyroid-hormone-binding protein (xCTBP), predominantly responsible for the major binding activity of T3 in the cytosol of Xenopus liver, has been shown to be identical to aldehyde dehydrogenase class 1 (ALDH1) [Yamauchi, K., Nakajima, J., Hayashi, H., Horiuchi, R. & Tata, J.R. A gene having high sequence homology (45–49%) with the glycerol-1-phosphate dehydrogenase gene from Methanobacterium thermoautotrophicum was cloned from the aerobic hyperthermophilic archaeon Aeropyrum pernix K1 (JCM 9820). This gene expressed in Escherichia coli with the pET vector system consists of 1113 nucleotides with an ATG initiation codon and a TAG termination codon. The Phycomyces blakesleeanus wild-type is yellow, because it accumulates b-carotene as the main carotenoid. A new carotenoid mutant of this fungus (A486) was isolated, after treatment with ethyl methane sulfonate (EMS), showing a whitish coloration. It accumulates large amounts of phytoene, small quantities of phyto¯uene, f-carotene and neurosporene, in decreasing amounts, and traces of b-carotene. Alcohol dehydrogenase (ADH) deficiency results in decreased retinol utilization, but it is unclear what physio-logical roles the several known ADHs play in retinoid signaling. Here, Adh1, Adh3,andAdh4null mutant mice have been examined following acute and chronic vitamin A excess. Following an acute dose of retinol (50 mgÆkg )1 ), metabolism of retinol to retinoic acid in liver was reduced 10-fold inAdh1mutants and 3.8-fold inAdh3mutants, but was not significantly reduced inAdh4mutants.

To gain better insight into the insulin secretory activity of fetalbcells in response to glucose, the expression of glucose transporter 2 (GLUT-2), glucokinase and mitochondrial glycerol phosphate dehydrogenase (mGDH) were studied. The present study reports the first purification and kinetic characterization of two plant arogenate dehydrogenases (EC 1.3.1.43), an enzyme that catalyses the oxidative decarboxylation of arogenate into tyrosine in presence of NADP. The twoArabidopsis thalianaarogenate dehy-drogenases TyrAAT1 and TyrAAT2 were overproduced in Escherichia coliand purified to homogeneity. Bio-chemical comparison of the two forms revealed that at low substrate concentration TyrAAT1 is four times more efficient in catalyzing the arogenate dehydrogenase reac-tion than TyrAAT2. ...

Short-chaindehydrogenases/reductases (SDRs) are enzymes of great functional diversity. Even at sequence identities of typically only 15–30%, specific sequence motifs are detect-able, reflecting common folding patterns. We have devel-oped a functional assignment scheme based on these motifs and we find five families. Two of these families were known previously and are calledclassical andextended families, but they are now distinguished at a further level based on coenzyme specificities.
