The two group I introns Nae.L1926 and Nmo.L2563, found at two different sites in nuclear LSU rRNA genes of Naegleria amoebo-flagellates, have been characterized in vitro. Their structural organization is related to that of the mobile Physarum intron Ppo.L1925 (PpLSU3) with ORFs extending the L1-loop of a typical group IC1 ribozyme. Nae.L1926, Nmo.L2563 and Ppo.L1925 RNAs all selfsplice in vitro, generating ligated exons and full-length intron circles as well as internal processed excised intron RNAs....

During starvation induced encystment, cells of the myxo-myceteDidymiumiridisaccumulate a7.5-kbRNAthat is the result of alternative processing of pre-rRNA. The 5¢ end corresponds to an internal processing site cleaved by the group I-like ribozyme DiGIR1, located within the twin-ribozyme intron Dir.S956-1. The RNAretains the majority ofDir.S956-1 including thehoming endonuclease geneanda small spliceosomal intron, the internal transcribed spacers ITS1 and ITS2, and the large subunit rRNAlacking its two group I introns. ...

The RuvABC resolvasome ofEscherichia colicatalyses the resolution of Holliday junctions that arise during genetic recombination and DNA repair. This process involves two key steps: branch migration, catalysed by the RuvB protein that is targeted to the Holliday junction by the structure specific RuvAprotein, and resolution, which is catalysed by the RuvC endonuclease.

Mammalian mitochondria contain strong nuclease activity. Endonuclease G (endoG), which predominantly resides in mitochondria, accounts for a large part of this nuclease activity. It has been proposed to act as an RNase H-like nuclease on RNAÆDNA hybrids (R-loops) in the D-loop region where the origins of mitochondrial replication are mapped, providing RNA primers for mtDNA replication.

The AnCOB group I intron fromAspergillus nidulans encodes a homing DNA endonuclease called I-AniI which also functions as a maturase, assisting in AnCOB intron RNA splicing. In this investigation we biochemically char-acterized the endonuclease activity of I-AniIin vitro and utilizedcompetitionassays toprobe the relationshipbetween theRNA- andDNA-binding sites.

Group I introns are thought to be self-propagating mobile elements, and are distributed over a wide range of organisms through horizontal trans-mission. Intron invasion is initiated through cleavage of a target DNA by a homing endonuclease encoded in an open reading frame (ORF) found within the intron. The intron is likely of no benefit to the host cell and is not maintained over time, leading to the accumulation of mutations after intron invasion.

Drosophila melanogasterXPG-like endonuclease (DmGEN) is a new cate-gory of nuclease belonging to the RAD2⁄XPG family. The DmGEN pro-tein has two nuclease domains (N and I domains) similar to XPG⁄class I nucleases; however, unlike class I nucleases, in DmGEN these two nuclease domains are positioned close to each other as in FEN-1⁄class II and EXO-1⁄ class III nucleases.

Adaptation to extreme environments affects the stability and catalytic effi-ciency of enzymes, often endowing them with great industrial potential. We compared the environmental adaptation of the secreted endonuclease I from the cold-adapted marine fish pathogenVibrio salmonicida(VsEndA) and the human pathogen Vibrio cholerae (VcEndA).

The genome of the human pathogenEntamoeba histolytica, a primitive pro-tist, contains non-long terminal repeat retrotransposable elements called EhLINEs. These encode reverse transcriptase and endonuclease required for retrotransposition. The endonuclease shows sequence similarity with bacterial restriction endonucleases.

Apurinic⁄apyrimidinic endonuclease (APE), an essential DNA repair enzyme, initiates the base excision repair pathway by creating a nick 5¢ to an abasic site in double-stranded DNA. Although the Chinese hamster ovary cells remain an important model for DNA repair studies, the Chinese hamster APE (chAPE1) has not been studied in vitro in respect to its kinetic characteristics.

Group II introns are both catalytic RNAs and mobile retroelements that move through a process catalyzed by a RNP complex consisting of an intron-encoded protein and the spliced intron lariat RNA. Group II intron-encoded proteins are multifunctional and contain an N-terminal reverse transcriptase domain, followed by a putative RNA-binding domain (domain X) associated with RNA splicing or maturase activity and a C-terminal DNA binding⁄DNA endonuclease region.
