The structural features of the hyperthermophilic endo-b-1,3-glucanase from Pyrococcus furiosuswere studied using circular dichroism, steady-state and time-resolved fluorescence spectroscopy and anisotropy. Upon heat and chemical treatment the folded and denatured states of the protein were characterized by distinguishable spectral profiles that identified a number of conformational states.

The induction of the heat shock response as well as its termination is auto-regulated by heat shock protein activities. In this study we have investi-gated whether Hsp90 functional protein levels influence the characteristics and duration of the heat shock response. Treatment of cells with several benzoquinone ansamycin inhibitors of Hsp90 (geldanamycin, herbimycin A) activated a heat shock response in the absence of heat shock, as reported previously.

Proteins tend to form inactive aggregates at high tempera-tures. We show that polyamines, which have a relatively simple structure as oligoamids, effectively prevent thermal inactivation and aggregation of hen egg lysozyme. In the presence of additives, including arginine and guanidine (100 mM), more than 30% of 0.2 mgÆmL )1 lysozyme in sodium phosphate buffer (pH 6.5) formed insoluble aggre-gates by heat treatment (98C for 30 min). 1

The tyrosine phenol-lyase fromSymbiobacterium toebiiwas engineered to improve both its stability and catalytic activity by the application of ran-dom mutagenesis and subsequent reassembly of the acquired mutations. Activity screening of the random library produced four mutants with a two-fold improved activity, whereas parallel screening after heat treatment at 65C identified three mutants with half-inactivation temperatures improved by up to 5.6 C.
