The current study presents the synthesis of thiophene-appended pyrazoles through 3+2 annulations of chalcones 3(a-g) with aryl hydrazine hydrochlorides 4(a-d) in acetic acid (30%) under reflux conditions produced the thiophene-pyrazole hybrids 5(a-g) in good yields.

Protein refolding is currently a fundamental problem in biophysics and molecular biology. We have studied the refolding process of frutalin, a tetrameric lectin that presents structural homology with jacalin but shows a more marked biological activity. The initial state in our refolding puzzle was that proteins were unfolded after thermal denaturation or denaturation induced by guanidine hydrochloride, and under both conditions, frutalin was refolded.

Duckd2-crystallin is a soluble tetrameric lens protein. In the presence of guanidinium hydrochloride (GdnHCl),it undergoes stepwise dissociation and unfolding. Gel-filtra-tion chromatography and sedimentation velocity analysis has demonstrated the dissociation of the tetramer protein to a monomeric intermediate with a dissociation constant of 0.34lM 3 . Dimers were also detected during the dissociation and refoldingprocesses.

Investigations of protein–solute interactions typically show that osmolytes favor native conformations. In this study, the effects of representative compatible and counteracting osmolytes on the reactivationof lactate dehydrogenase from two different conformational states were explored. Contrary to expectations, proline and trimethylamine oxide inhibited both the initial time course and the extent of reactivation of lactate dehydrogenase from bovine heart following dena-turation in guanidine hydrochloride, as well as following inactivation at pH 2.3.

To probe the stability of the seven-iron ferredoxin from Thermus thermophilus(FdTt), we investigated its chemical and thermal denaturationprocesses insolution.Aspredicted from the crystal structure, FdTt is extremely resistant to perturbation. The guanidine hydrochloride-induced unfolding transition shows a midpoint at 6.5M(pH 7, 20C), andthe thermalmidpoint is aboveboiling, at 114C. The stability of FdTt is much lower at acidic pH, suggesting that electrostatic interactions are important for the high stability at higher pH.

The CII protein of bacteriophagek, which activates the synthesis of thekrepressor, plays a key role in the lysis– lysogeny switch. CII has a small in vivo half-life due to its proteolytic susceptibility, and this instability is a key com-ponent for its regulatory role. The structural basis of this instability is not known. While studying guanidine hydro-chloride-assisted unfolding of CII, we found that low concentrations of the chaotrope (50–500 mM) have a con-siderable effect on the structure of this protein.

Thermodynamic parameters describing the conformational stability of the histidine-containing phosphocarrier protein fromStreptomyces coelicolor, scHPr, have been determined by steady-state fluorescence measurements of isothermal urea-denaturations, differential scanning calorimetry at different guanidinium hydrochloride concentrations and, independently, by far-UV circular dichroism measurements of isothermal urea-denaturations, and thermal denatura-tions at fixed urea concentrations.

The unfolding pathways of the two forms ofPlasmodium falciparumacyl carrier protein, the apo and holo forms, were determined by guanidine hydrochloride-induced denaturation. Both the apo form and the holo form displayed a reversible two-state unfolding mechanism. The analysis of iso-thermal denaturation data provides values for the conformational stability of the two proteins.

Onconase is an extremely stable member of the RNase A superfamily. The increase in the thermodynamic stability by 20 kJÆmol )1 in comparison to RNase A was expected to result in altered folding behavior. Despite the lack of cis-Pro residues in native Onconase, refolding at low concentrations of guanidine hydrochloride was complex and showed three kinetic phases (fast, medium, and slow), with rate constants differing by a factor of about 10 each.
