4-Coumarate:CoA ligase (4CL) is involved in the formation of coenzyme A thioesters of hydroxycinnamic acids that are central substrates for subsequent condensation, reduction, and transfer reactions in the biosynthesis of plant phenylpropanoids. Previous studies of 4CL appear to suggest that many isoenzymes are functionally equivalent in supplying substrates to various subsequent branches of phenylpropanoid biosyntheses. In contrast, divergent members of a 4CL gene family were identified in soybean (Glycine max L.).

Murine double minute 2 (MDM2) protein exhibits many diverse biochemi-cal functions on the tumour suppressor protein p53, including transcrip-tional suppression and E3 ubiquitin ligase activity. However, more recent data have shown that MDM2 can exhibit ATP-dependent molecular chap-erone activity and directly mediate folding of the p53 tetramer. Analysing the ATP-dependent function of MDM2 will provide novel insights into the evolution and function of the protein.

A novel plasmid vector pSELECT-1 is described which can be used for highly efficient site-directed in vitro mutagenesis. The mutagenesis method is based on the use of single-stranded DNA and two primers, one mutagenic primer and a second correction primer which corrects a defect in the ampicillin resistance gene on the vector and reverts the vector to ampicillin resistance. Using T4 DNA polymerase and T4 DNA ligase the two primers are physically linked on the template. The non-mutant DNA strand is selected against by growth in the presence of ampicillin.

Biotin protein ligase (BPL) catalyses the biotinylation of the biotin carboxyl carrier protein (BCCP) subunit of acetyl CoAcarboxylase and this post-translational modificationof a single lysine residue is exceptionally specific. The exact details of the protein–protein interactions involved are unclear as a BPL:BCCP complex has not yet been isolated. Moreover, detailed information is lacking on the composi-tion, biosynthesis and role of fatty acids in hyperthermo-philic organisms.

The surface charge as well as the electrochemical properties and ligand binding abilities of the Gram-positive cell wall is controlled by the d-alanylation of the lipoteichoic acid. The incorporation of d-Ala into lipoteichoic acid requires the d-alanine:d-alanyl carrier protein ligase (DltA) and the carrier protein (DltC). We have heterologously expressed, purified, and assayed the substrate selectivity of the recombinant proteins DltA with its substrate DltC.

The suppressors of cytokine signalling (SOCS) proteins inhibit cytokine signalling by direct interaction with Janus kinases (JAKs) or activated cytokine receptors. In addition to the N-ter-minal and SH2 domains that mediate these interactions, SOCS proteins contain a C-terminal SOCS box. Evidence suggests SOCS box-containing proteins act as part of an elongin-cullin-SOCS (ECS) E3 ubiquitin ligase complex, marking target pro-teins for degradation.
