In contrast with traditional nuclear gene transformation, transplastomic technology has opened a new horizon for the transgenic plant research that offers several beneficial aspects including the convenient use of transgene stacking and the generation of high expression levels of foreign proteins. However, this technology has been well adopted and established in tobacco, the introduction and adoption of cost-effective, swift, and reproducible protocol for in vitro regeneration of transplastomic potato is challenging and laborious.

Potato (Solanum tuberosum L.) growth, development and production are sensitive to abiotic stress conditions, yet studies about investigation of stress response of potato plants in regulation of gene expression level are limited. The aim of this study was to identify the role of newly identified Novel_105 miRNA in contrasting potato cultivars (tolerant Unica and sensitive Russet Burbank) in response to heat, drought and combined heat and drought stresses using transgenic approach.

The presence of bov and ZRNase II transgenes was confirmed by PCR analysis. RNase activity was examined by modified Oleshko method. Plants with heterologous ribonuclease genes had higher level of RNase activity compared to nontransgenic ones. Transgenic plants inoculated with Potato virus Y, PVY (genus Potyvirus, family Potyviridae) demonstrated delayed and less severe symptoms of viral infection. DAS-ELISA confirmed the presence of viral antigens both in transformed and control plants.

Common scab caused by Streptomyces scabies (Thaxter) Waksman & Henrici is an important disease worldwide, and can cause significant reduction in the economic value of potato. The management of the disease can be done through conventional methods, biological chemical and cultivation of resistant variety. However, resistance against pathogen is not a permanent solution and fungicidal is not ecofriendly and economical. But new search, induce resistance has taken great attention for plant disease management in near future.
