Two genes (CM-AAT1 and CM-AAT2) with strong sequence homology (87% identity at the protein level) putatively involved in the formation of aroma volatile esters have been isolated from Charentais melon fruit. They belong to a large and highly divergent family of multifunctional plant acyl-transferases and show at most 21% identity to the only other fruit acyl-transferase characterized so far in strawberry. RT-PCR studies indicated that both genes were specifically expressed in fruit at increasing rates in the early and mid phases of ripening.

Binding of lamin B receptor (LBR) to chromatin was studied by means of an in vitro assay system involving recombinant fragments of human LBR and Xenopus sperm chromatin. Glutathione-S-transferase (GST)-fused proteins including LBR fragments containing the N-terminal region (residues 1–53) and arginine-serine repeat-containing region (residues 54–89) bound to chromatin. The binding of GST-fusion proteins incorporating the N-terminal and arginine-serine repeat-containing regions to chromatin was suppressed by mild trypsinization of the chromatin and by pretreatment with a DNA solution....

Antisense oligodeoxynucleotides (AS-ODNs) are frequently used for the down-regulation of protein expression.Because the majority of potential antisense sequences lacks effect-iveness, fast screening methods for the selection of effective AS-ODNs are needed. We describe a new cellular screening assay for the evaluationof the potency and specificityof new antisense sequences.

Propionate CoA-transferase fromClostridium propionicum hasbeenpuri®edand thegene encoding the enzymehasbeen cloned and sequenced. The enzyme was rapidly and irre-versibly inactivated by sodium borohydride or hydroxyl-amine in the presence of propionyl-CoA. The reduction of the thiol ester between a catalytic site glutamate and CoA with borohydride and the cleavage by hydroxylamine were used to introduce a site-speci®c label, which was followed by MALDI-TOF-MS. This allowed the identi®cation of glutamate 324 at the active site. ...

b1,4-N-Acetylglucosaminyltransferase III (GnT-III) plays a regulatory role in the biosynthesis of N-glycans, and it has been suggested that its product, a bisecting GlcNAc, is involved in a variety of biological events as well as in regu-lating the biosynthesis of the oligosaccharides. In this study, it was found, on the basis of sequence homology, that GnT-III contains a small region that is signi®cantly homologous to both snail b1,4GlcNAc transferase andb1,4Gal trans-ferase-1.

This paper deals with the purification and the partial char-acterization of glutathione S-transferase (GST) isoforms from the clam Ruditapes decussatus. For the first step of purification, two affinity columns, reduced glutathione (GSH)–agarose and S-hexyl GSH–agarose, were mounted in series. Four affinity fractionswere thus recovered. Further purification was performed using anion exchange chroma-tography. Seven fractions, whichpresent aGSTactivitywith 1-chloro-2,4-dinitrobenzene (CDNB) as substrate, were collected and analyzed by RP-HPLC....

Biosynthesis ofmucin-typeO-glycans is initiated by a family of UDP-GalNAc:polypeptide N-acetylgalactosaminyl-transferases, which contain several conserved cysteine resi-dues among the isozymes. We found that a cysteine-specific reagent, p-chloromercuriphenylsulfonic acid (PCMPS), irreversibly inhibited one of the isozymes (GalNAc-T1). Presence of either UDP-GalNAc or UDP during PCMPS treatment protected GalNAc-T1 from inactivation, to the same extent.

Partially glycosylated versions of this peptide, PT*TTPLK, PTTT*PLK, PT*TT*PLK, PTT*T*PLK, PT*TTPLK, andPTTT*PLK(*,N-acetyl-galactosamine;, galactose), were also tested.

GlutathioneS-transferases (EC 2.5.1.18) (GSTs), are a family of multifunctional enzymes present in all living organisms whose main function is the detoxification of electrophilic compounds. GSTs are considered the most prominent detoxifying class II enzymes in helminths. We describehere the characterizationof novel dehydroascorbate reductase and thiol transferase activities that reside in the human parasite Schistosoma mansoni GSTx.

Thirteen glucose analogues bearing electrophilic groups were synthesized (five of themfor the first time) and screened as inhibitors of the glucose transporter (EII Glc )of the Escherichia coli phosphoenolpyruvate–sugar phospho-transferase system (PTS). 2¢,3¢-Epoxypropyl b-D-glucopyr-anoside (3a) is an inhibitor and also a pseudosubstrate. Five analogues are inhibitorsofnonvectorialGlcphosphorylation by EII Glc but not pseudosubstrates.

Rubber transferase, acis-prenyltransferase, catalyzes the addition of thousands of isopentenyl diphosphate (IPP) molecules toanallylic diphosphate initiator, suchas farnesyl diphosphate (FPP,1), in the presence of a divalent metal cofactor. In an effort to characterize the catalytic site of rubber transferase, the effects of two types of protein farnesyltransferase inhibitors, several chaetomellic acid A analogs (2,4–7)anda-hydroxyfarnesylphosphonic acid (3), on the ability of rubber transferase to add IPP to the allylic diphosphate initiator were determined. ...

The a-amylase family (glycoside hydrolase family 13; GH 13) contains enzymes with approximately 30 specifi-cities. Six types of enzyme from the family can possess a C-terminal starch-binding domain (SBD): a-amylase, maltotetraohydrolase, maltopentaohydrolase, maltogenic a-amylase, acarviose transferase, and cyclodextrin glu-canotransferase (CGTase). Such enzymes are multidomain proteins and those that contain an SBD consist of four or five domains, the former enzymes being mainly hydrolases and the latter mainly transglycosidases. ...
