Gluconobacter oxydans is a strictly aerobic Gram-negative acetic acid bacterium used industrially for oxidative biotransformations due to its exceptional type of catabolism. It incompletely oxidizes a wide variety of carbohydrates regio- and stereoselectively in the periplasm using membrane-bound dehydrogenases with accumulation of the products in the medium.

Morphological evaluation of embryos has been used to screen embryos for transfer. However, the repeatability and accuracy of this method remains low. Thus, evaluation of an embryo’s gene expression signature with respect to its developmental capacity could provide new opportunities for embryo selection.

The interaction between the c11ring and thececomplex, forming the rotor of theIlyobacter tartaricus ATP synthase, was probed by surface plasmon resonance spectroscopy andin vitro reconstitution analysis. The results pro-vide, for the first time, a direct and quantitative assessment of the stability of the rotor. The data indicated very tight binding between the c11ring and the cecomplex, with an apparent Kd value of approximately 7.4 nm.

The subunit c from the ATP synthase ofPropionigenium modestumwas studied by NMR in chloroform/methanol/ water (4 : 4 : 1). In this solvent, subunit c consists of two helical segments, comprisedof residuesL5 to I26andG29 to N82, respectively. On comparing the secondary structure of subunit c fromP. modestumin the organic solvent mixture with that in dodecylsulfate micelles several deviations became apparent: in the organic solvent, the interruption of theahelical structure within the conserved GXGXGXGX motifwas shortened from®ve to tworesidues, ...

The interface between thec-subunit oligomer and the asubunit in the F0sector of the ATP synthase is believed to formthe core of the rotatingmotor powered by the protonic ¯ow. Besides the essential cAsp61 and aArg210 residues (Escherichia colinumbering), a few other residues at this interface, although nonessential, show a high degree of conservation, among these aGlu219.

Recent crosslinking studies indicated the localization of the coupling ion binding site in the Na + -translocating F1F0 ATP synthase ofIlyobacter tartaricus within the hydrophobic part of the bilayer.Similarly, a membrane embedded H + -binding site is accepted for the H + -trans-locating F1F0 ATP synthase ofEscherichia coli. For a more definite analysis, we performed parallax analysis of fluorescence quenching with ATP synthases from both I. tartaricus and E. coli.

Cytidine 5¢-triphosphate synthase catalyses the ATP-dependent formation of CTP from UTP using either ammonia orL-glutamine as the source of nitrogen. When glutamine is the substrate, GTP is required as an allosteric effector to promote catalysis. Limited trypsin-catalysed proteolysis, Edman degradation, and site-directed muta-genesis were used to identify peptide bonds C-terminal to three basic residues (Lys187, Arg429, and Lys432) of Escherichia coliCTP synthase thatwere highly susceptible to proteolysis. ...

A conserved putative dimerization GxxxG motif located in the unique membrane-spanning segment of the ATP syn-thase subunit e was altered in yeast both by insertion of an alanine residue and by replacement of glycine by leucine residues. These alterations led to the loss of subunit gand the loss of dimeric and oligomeric forms of the yeast ATP syn-thase. Furthermore,as in null mutants devoid of either subunit e or subunit g,mitochondria displayed anomalous morphologies with onion-like structures. ...

Depolarization and repolarization phases (D and R phases, respectively) of mitochondrial potential fluctuations induced by photoactivation of the fluorescent probe tetramethylrhodamine methyl ester (TMRM) were ana-lyzed separately and investigated using specific inhibitors and substrates. The frequency of R phases was significantly inhibited by oligomycin and aurovertin (mitochondrial ATP synthase inhibitors), rotenone (mitochond-rial complex I inhibitor) and iodoacetic acid (inhibitor of the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase)....

The recombinantc genes were overexpressed inEscherichia coliand assembled withabsub-units into a native complex. The wild-type (WT)abcassembly i.e.abcWT exhibited high Mg 2+ -dependent and Ca 2+ -dependent ATP hydrolytic activity.

Cytidine 5¢-triphosphate (CTP) synthase catalyses the ATP-dependent formation of CTP from uridine 5¢-triphosphate using either NH3orL-glutamine as the nitrogen source. The hydrolysis of glutamine is catalysed in the C-terminal glu-tamine amide transfer domain and the nascent NH3that is generated is transferred via an NH3 tunnel [Endrizzi, J.A., Kim, H., Anderson, P.M. & Baldwin, E.P. (2004) Biochemistry43, 6447–6463] to the active site of the N-ter-minal synthasedomainwhere theaminationreactionoccurs....

Mitochondrial biogenesis is a complex and highly regulated process that requires the controlled expression of hundreds of genes encoded in two separated genomes, namely the nuclear and mitochondrial genomes. To identify regulatory proteins involved in the transcriptional control of key nuc-lear-encoded mitochondrial genes, we have performed a detailed analysis of the promoter region of theasubunit of the Drosophila melanogasterF1F0 ATP synthase complex.

Acentral shaft, subunitc, functionally connects the ion-driven rotary motor, FO, with the rotary chemical reactor, F1.Using polarized spectrophotometry we have demonstrated previ-ously the functional rotation of the C-terminala-helical portion ofcin the supposed ‘hydrophobic bearing’ formed by the (ab)3hexagon. In apparent contradiction with these spectroscopic results, an engineered disulfide bridge between thea-helixofcand subunitadidnot impair enzyme activity....

Heat shock inducible lysyl-tRNA synthetase ofEscherichia coli(LysU) is known to be a highly efficient diadenosine 5¢,5¢¢¢-P 1 ,P 4 -tetraphosphate (Ap4A) synthase. However, we use an ion-exchange HPLC technique to demonstrate that active LysU mixtures actually have a dual catalytic activ-ity, initially producing Ap4 A from ATP, before converting that tetraphos-phate to a triphosphate.
