Keeping poultry makes a substantial contribution to household food security throughout the developing world. It helps diversify incomes and provides quality food, energy, fertilizer and a renewable asset in over 80 percent of rural households. Small-scale producers are however constrained by poor access to markets, goods and services; they have weak institutions and lack skills, knowledge and appropriate technologies. The result is that both production and productivity remain well below potential and losses and wastage can be high.

Reactionof certainpeptides andproteinswithsingletoxygen (generated by visible light in the presence of rose bengal dye) yields long-lived peptide and protein peroxides. Incubation of these peroxides with glyceraldehyde-3-phosphate dehy-drogenase, in the absence of addedmetal ions, results in loss of enzymatic activity. Comparative studies with a range of peroxides have shown that this inhibition is concentration, peroxide, and time dependent, with H2O2less ecient than some peptide peroxides.

In this study we investigate the active-site structure and the catalytic mechanism of clostripain by using a combination of three separate techniques: affinity labelling, site-directed mutagenesis and molecular modelling. A benzamidinyl-diazo dichlorotriazine dye (BDD) was shown to act as an efficient active site-directed affinity label for Clostridium histolyticumclostripain. The enzyme, upon incubation with BDD in 0.1MHepes/NaOHbuffer pH 7.6, exhibits a time-dependent loss of activity.

Metronidazole (MTZ) is widely used in combination therapies against the human gastric pathogenHelicobacter pylori. Resistance to this drug is com-mon among clinical isolates and results from loss-of-function mutations in rdxA, which encodes an oxygen-insensitive nitroreductase. The RdxA-associated MTZ-reductase activity ofH. pyloriis lost upon cell disruption. Here we provide a mechanistic explanation for this phenomenon.
