In avian species, an egg envelope homologous to the mammalian zona pellucida is called the perivitelline membrane. We have previously reported that one of its components, a glycoprotein homologous to mammalian ZPC, is synthesized in the granulosa cells of the quail ovary. In the present study, we investigated the proteolytic cleavage of the newly synthesized ZPC and the secretion of ZPC from the granulosa cells. Western blot analysis of the cell lysates demonstrated that the 43-kDa protein is the precursor of mature ZPC (proZPC), and is converted to the 35-kDa protein before secretion. ...

Thioredoxin is a ubiquitous dithiol oxidoreductase found in many organisms and involved in numerous biochemical processes. Human thioredoxin-like protein (hTRXL) is differentially expressed at different development stages of human fetal cerebrum and belongs to an expanding family of thioredoxins. We have solved the crystal structure of the recombinant N-terminal catalytic domain (hTRXL-N) of ˚ hTRXL in its oxidized form at 2.2-A resolution.

InHIV the viral envelope protein is processed by a host cell protease toformgp120andgp41.TheC1andC5domainsof gp120 are thought to directly interact with gp41 but are largely missing from the available X-ray structure. Bio-physical studies of theHIVgp120C5 domain (residues 489– 511 of HIV-1 strain HXB2), which corresponds to the carboxy terminal region of gp120, have been undertaken.

Arecombinant streptococcalC5apeptidasewas expressed in Escherichia coliand its catalytic properties and thermal sta-bility were subjected to examination. It was shown that the NH2 -terminal region ofC5a peptidase (Asn32–Asp79/ Lys90) forms the pro-sequence segment. Upon maturation the propeptide is hydrolyzed either via an autocatalytic intramolecular cleavage or by exogenous protease strepto-pain. At pH 7.4 the enzyme exhibited maximum activity in the narrow range oftemperatures between 40 and 43C....

Biosynthesis of cholesterol represents one of the funda-mental cellular metabolic processes. SterolD14-reductase (D14-SR) is a microsomal enzyme involved in the con-version of lanosterol to cholesterol in mammals. Amino-acid sequence analysis of a 38-kDa protein puri®ed from bovine liver in our laboratory revealed 90% similarity with a human sterol reductase, SR-1, encoded by the TM7SF2gene, and with the C-terminal domain of human lamin B receptor.

Transglutaminase (TGase) fromStreptomyces mobaraensis is secreted as a precursor protein which is completely acti-vated by the endoprotease TAMEP, a member of the M4 protease family [Zotzel, J., Keller, P. & Fuchsbauer, H.-L. (2003) Eur. J. Biochem. 270, 3214–3222]. In contrast with the mature enzyme, TAMEP-activated TGase exhibits an additional N-terminal tetrapeptide (Phe-Arg-Ala-Pro) sug-gesting truncation, at least, by a second protease.

In the leukemic macrophage cell-line THP-1, a fraction of the secreted matrix metalloproteinase 9 (MMP-9) is linked to the core protein of chondroitin sulfate proteoglycans (CSPG). Unlike the monomeric and homodimeric forms of MMP-9, the addition of exogenous CaCl2 to the proMMP-9/CSPG complex resulted in an active gelatinase due to the induction of an autocatalytic removal of the N-terminal prodomain.

Protein phosphatase 2A is a phosphoserine/threonine phosphatase implicated inmany cellular processes. The core enzyme comprises a catalytic and a PR65/A-subunit. The substrate specificity and subcellular localization are deter-mined by a third regulatory B-subunit (PR55/B, PR61/B¢ and PR72/130/B¢¢). To identify the proteins of the B¢¢family inXenopus laevisoocytes, aprophaseXenopusoocyte cDNA librarywas screened using humanPR130 cDNAas a probe. Three different classes of cDNAs were isolated....

Topogenesis of cytochrome P450scc, a resident proteinof the inner membrane of adrenocortical mitochondria, is still obscure. In particular, little is known about the cause of its tissue specificity. In an attempt to clarify this point, we examined the process in Saccharomyces cerevisiae cells synthesizing cytochrome P450scc as its native precursor (pCYP11A1) or versions inwhich its N-terminal addressing presequence had been replaced with those of yeast mitoch-ondrial proteins: CoxIV(1–25) and Su9(1–112)....

Calcium metabolism in oysters is a very complicated and highly controlled physiological and biochemical process. However, the regulation of calcium metabolism in oyster is poorly understood. Our previous study showed that calmodulin (CaM) seemed to play a regulatory role in the process of oyster calcium metabolism. In this study, a full-length cDNA encoding a novel calmodulin-like protein (CaLP) with a long C-terminal sequence was identi-fied from pearl oysterPinctada fucata, expressed in Escherichia coliand characterizedin vitro. ...

The transporter associated with antigen processing (TAP) delivers peptides into the lumen of the endoplasmic reticu-lum for binding onto major histocompatibility complex class I molecules. TAP comprises two polypeptides,TAP1 andTAP2,eachwithanN-terminal transmembranedomain and a C-terminal cytosolic nucleotide binding domain (NBD). The two NBDs have distinct intrinsic nucleotide binding properties.

Phospholipase D1 (PLD1) is a signal-transduction regulated enzyme which regulates several cell intrinsic processes including activation of NAPDH oxidase, which elevates intracellular H2O2. Several proteins have been reported to interact with PLD1 in resting cells. We sought to identify pro-teins that interact with PLD1 after phorbol 12-myristate 13-acetate (PMA) stimulation.

The highly conserved, atypical RIO serine protein kinases are found in all organisms, from archaea to man. In yeast, the kinase activity of Rio2 is necessary for the final processing step of maturing the 18S ribosomal rRNA. We have previously shown that the Rio2 protein from Archaeo-globus fulgidus contains both a small kinase domain and an N-terminal winged helix domain.

The gene encoding an integrase of Mason–Pfizer monkey virus (M-PMV) is located at the 3¢-end of the pol open reading frame. The M-PMV integ-rase has not been previously isolated and characterized. We have now cloned, expressed, isolated, and characterized M-PMV integrase and com-pared its activities and primary structure with those of HIV-1 and other retroviral integrases. M-PMV integrase prefers untranslated 3¢-region-derived long-terminal repeat sequences in both the 3¢-processing and the strand transfer activity assays.

Purified, nonubiquitinated growth-associated protein of 43 kDa (GAP-43) was attacked by purified reticulocyte 20S proteasomebutnotby the26Sproteasome.Cleavageyielded 12 N-terminally labelled GAP-43 fragments that could be resolved by SDS/PAGE. Inhibitor experiments suggested that proteasomeb1 activity yielded the resolved bands and that proteasome b5 activity generated nonresolvable frag-ments. Processive degradation, yielding only nonresolvable fragments, therefore did not occur.

Onconase, a member of the ribonuclease superfamily, is a potent cytotoxicagent that isundergoingphase II/IIIhuman clinical trials as an antitumor drug. Native onconase from Rana pipiensand its amphibian homologs have an N-ter-minal pyroglutamyl residue that is essential for obtaining fully active enzymes with their full potential as cytotoxins. When expressed cytosolically in bacteria, Onconase is isolated with an additional methionyl (Met1) residue and glutaminyl insteadof a pyroglutamyl residue at position 1 of the N-terminus and is consequently inactivated....
